Article Abstract

Charge detection mass spectrometry (CDMS) is a well-established technique that provides direct mass spectral outputs regardless of analyte heterogeneity or molecular weight. Over the past few years, it has been demonstrated that CDMS can be multiplexed on Orbitrap analyzers utilizing an integrated approach termed individual ion mass spectrometry (IMS). To further increase adaptability, robustness, and throughput of this technique, here, we present a method that utilizes numerous integrated equipment components including a Kingfisher system, SampleStream platform, and Q Exactive mass spectrometer to provide a fully automated workflow for immunoprecipitation, sample preparation, injection, and subsequent IMS acquisition. This automated workflow has been applied to a cohort of 58 test subjects to determine individualized patient antibody responses to SARS-CoV-2 antigens. Results from a range of serum donors include 37 subject IMS spectra that contained a positive COVID-19 antibody response and 21 IMS spectra that contained a negative COVID-19 antibody response. This high-throughput automated IMS workflow can currently process over 100 samples per week and is general for making immunoprecipitation-MS workflows achieve proteoform resolution.

B-lymphocytes play major adaptive immune roles, producing antibody and driving T-cell responses. However, how immunometabolism networks support B-cell activation and differentiation in response to distinct receptor stimuli remains incompletely understood. To gain insights, we systematically investigated acute primary human B-cell transcriptional, translational and metabolomic responses to B-cell receptor (BCR), Toll-like receptor 9 (TLR9), CD40-ligand (CD40L), interleukin-4 (IL4) or combinations thereof.

The strong C-F bond found in per- and poly-fluorinated alkyl substances (PFAS) makes them resistant to degradation and thus persistent in the environment. One of the most common methods for quantifying PFAS in environmental matrices is to use tandem mass spectrometry. However, the dissociation of ions made by deprotonating PFAS alcohols and acids has only been qualitatively explored.

A method for determination of ten kinds of sweeteners in soybean products by multi-plug filtration cleanup (-PFC) combined with ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was established. The sample was extracted with acetonitrile (containing 1% formic acid), degreased by using -hexane liquid-liquid extraction and purified by solid phase extraction using an -PFC column (Oasis PRiME HLB). The analytes were separated by using a Waters ACQUITY UPLC® BEH C (2.

The purpose of this study was to evaluate the pharmacokinetics of oral (PO) ondansetron compared to intravenous (IV) ondansetron in eight healthy client-owned dogs. Dogs were randomized to one of two protocols in a crossover design, receiving PO or IV ondansetron at a dose of 1 mg/kg on Day 0 and the opposite formulation at an equal dose on Day 7. Plasma was collected at baseline and 1, 2, 4, and 8 h post administration.
