Hepatocyte nuclear factor 4 alpha (Hnf4α), a conserved nuclear receptor central to vertebrate liver development and metabolic regulation, emerges here as a pivotal immune regulator in teleosts against complex infectious threats. While its metabolic roles are well-established, Hnf4α's function in bacterial infection, viral infection, and bacterial-viral coinfection-major challenges in global aquaculture-remained uncharacterized. This study reveals that teleost Hnf4α acts as a dual-functional immune checkpoint, essential for combating Aeromonas salmonicida, grass carp reovirus (GCRV), and their coinfection.

Background: Memory CD8 T cells sense inflammation and rapidly produce interferon-γ (IFN-γ) independent of cognate antigens. This innate-like property, called bystander activation, is involved in early host defense before the antigen-specific memory response. However, the molecular mechanisms underlying this activation remain unknown.

A20, encoded by the TNFAIP3 gene, is a protein linked to Crohn's disease and celiac disease in humans. We now find that mice expressing point mutations in A20's M1-ubiquitin-binding zinc finger 7 (ZF7) motif spontaneously develop proximal enteritis that requires both luminal microbes and T cells. Cellular and transcriptomic profiling reveals expansion of Th17 cells and exuberant expression of IL-17A and IL-22 in intestinal lamina propria of A20ZF7 mice.

Polydatin is a bioactive compound extracted from the roots of Reynoutria japonica Houtt. It has demonstrated various protective effects on the cardiovascular system. However, the underlying molecular mechanisms through which polydatin exerts its effects in atherosclerosis (AS) remain largely unclear, particularly regarding the involvement of the ferroptosis pathway in its anti-atherosclerotic action.

Macrophage extracellular traps (METs) are crucial for initiating airway inflammation and modulating the immune microenvironment of asthmatic airways. Metrnl/IL-41 is a negative regulator of airway inflammatory responses. However, the role of Metrnl/IL-41 in the cross-talk between METs and airway epithelial cells, as well as its effect on the pathophysiology of asthma airway remodeling, are still unclear.
