Article Abstract

Introduction: Acute myeloid leukemia with normal cytogenetics (CN-AML) represents a heterogeneous group having diverse genetic mutations. Understanding the significance of each of these mutations is necessary. In this study, we evaluated the prognostic role of MN1 expression in adult CN-AML patients.

Method: One hundred and sixty-three de-novo adult AML patients were evaluated for MN1 expression by real-time PCR. MN1 expression was correlated with the clinical characteristics of the patients and their outcomes.

Results: Higher MN1 expression was associated with NPM1 wild-type (p<0.0001), CD34 positivity (p=0.006), and lower clinical remission rate (p=0.027). FLT3-ITD and CEBPA mutations had no association with MN1 expression. On survival analysis, a high MN1 expression was associated with poor event-free survival (Hazard Ratio 2.47, 95% Confidence Interval: 1.42-4.3; p<0.0001) and overall survival (Hazard Ratio 4.18, 95% Confidence Interval: 2.17-8.08; p<0.0001). On multivariate analysis, the MN1 copy number emerged as an independent predictor of EFS (p<0.0001) and OS (p<0.0001).

Conclusion: MN1 expression is an independent predictor of outcome in CN-AML.

Corneal alkali-burn injury is a common ophthalmic emergency that elevates oxidative stress in the ocular microenvironment, leading to cellular apoptosis and corneal neovascularization. To address this challenge, we report a binuclear manganese complex () featuring a catalase-mimetic antiferromagnetic Mn-O-Mn core, which efficiently scavenges hydrogen peroxide, a key contributor to oxidative damage. This structure enables effective hydrogen peroxide scavenging while concurrently suppressing the Fenton reaction, a major driver of oxidative injury.

GDNF is a potent survival and differentiation factor for motor neurons and other central and peripheral neuronal populations. While the signaling pathways by which GDNF promotes survival/differentiation have been relatively well established, the molecular mechanisms that restrict its biological effects remain unclear. In this study, we show that TDAG51 plays a role in regulating the GDNF-induced PI3K/AKT survival pathway.

Canonical MN1 fusions with either BEND2 or CXXC5 gene partners represent the molecular hallmark of astroblastoma, a stand-alone group among central nervous system (CNS) high-grade neuroepithelial tumors based on their distinct methylation profile. Outside the CNS, MN1 fusions have been rarely reported, mostly with nonrecurrent gene partners. Herein, we present three cases of soft tissue sarcomas harboring MN1 gene rearrangements, two of which had MN1 (exon 1)::CXXC5 (exon 2) gene fusion and the last had MN1 (exon 1)::ZFP64 (exon 2) gene fusion.

Low-Grade Primary Intramedullary Spinal Cord Astroblastoma: A Case Report and Literature Review.

Spinal astroblastoma is an exceedingly rare entity characterized by features that overlap with other spinal cord tumors. We present a case of a 67-year-old male who presented with trunk dysesthesia, motor weakness, and progressive hypoesthesia in both lower limbs. Magnetic resonance imaging (MRI) revealed edematous changes in the spinal cord at the C6-Th1 level on T2-weighted sequences, with a centrally enhancing lesion at the C7 level on gadolinium-enhanced T1-weighted imaging.
