Article Abstract

A small library of easily prepared diamidophosphite-sulfides based on 1,3-thioether alcohols, primarily of terpenoid nature, was developed. Upon complexation with Pd(II) ions, these hemilabile ligands showed the ability to form both ,-chelates and complexes with two ligands -monodentately bonded to the metal. The structures of the ligands and their complexes were determined by 2D NMR spectroscopy and X-ray diffraction. The use of these stereoselectors provided up to 95% ee in the classic Pd-catalyzed asymmetric allylic substitution reactions of ()-1,3-diphenylallyl acetate with C- and N-nucleophiles and up to 80% ee in the Pd-mediated allylic alkylation of cinnamyl acetate with β-ketoesters. In addition, ee values of up to 90% with quantitative conversion were achieved in the Rh-catalyzed asymmetric hydrogenation of methyl esters of unsaturated acids. The effects of the structural parameters, reaction conditions and ligand-to-metal ratio on the catalytic results are discussed.

Based on a patented new concept of general chiral catalysis, a series of new chiral ligands integrating both a MacMillan imidazolinone organocatalyst and a chiral N,P-ligand scaffold has been designed and concisely prepared from commercially available chiral α-amino acids and 2-diphenylphosphino-benzaldehyde. These chiral ligands have shown good to excellent enantioselectivities (up to 97% ee) with satisfactory yields (up to 95%) in enantioselective Pd-catalyzed allylations with 1,3-dicarbonyls and amines.

We synthesized a series of phosphine-acrylamide chiral compounds 4 from chiral aminophosphine ()-5 with various acyl chlorides and demonstrated that they can be used as chiral ligands in the Pd-catalyzed asymmetric allylic substitution reactions of allylic acetate with malonates or indoles. Although the ligands shared the same axial chirality, we found that the substitution pattern of the acrylamide led to an inversion in the stereoselectivity of the product and then investigated the underlying mechanism based on DFT calculations.

Chiral γ-lactam scaffolds represent a privileged structural motif that is prominently featured in numerous biologically active natural products and pharmaceuticals. Notably, due to the intimidating challenges in the stereocontrol for the intramolecular amidation, the asymmetric cascade carboamidation of the C═C bond to access structurally diverse chiral γ-lactams from readily available 2-bromoamides remains unexplored. Pd-catalyzed asymmetric (6+2) dipolar cyclization for medium-sized spirooxindoles.

Chiral spirooxindoles are privileged scaffolds in bioactive molecules, yet efficient strategies for their functionalization-especially for medium-sized spirocycles-remain scarce. We report a Pd-catalyzed asymmetric (6+2) dipolar cyclization between vinyloxetanes and 3-diazoquinoline-2,4-diones, which allows the synthesis of enantioenriched spirooxindoles with eight-membered lactones under mild conditions. This method provides 22 examples with high yields and enantioselectivities (up to 90% yield and 98% ee), facilitated by a tailored chiral phosphine ligand and photogenerated ketene dipolarophiles.

Chiral allenyl phosphine oxides hold significant application value in organic synthesis, serving as efficient catalysts, ligands, and versatile synthons. However, the development of transition metal-catalyzed asymmetric synthesis for these compounds remains unexplored, primarily due to the facile tautomerization of phosphine oxides into trivalent RRP-OH species, which may severely deactivate transition metal catalysts through strong coordination interactions. Herein, a ligand relay strategy has been applied to address this challenge in the Pd-catalyzed enantioselective coupling reactions of propargylic benzoates and secondary phosphine oxides (SPOs) to afford chiral trisubstituted allenyl phosphine oxides with high ee.
