The parathyroid hormone receptor type 1 (PTH1R) is a G protein-coupled receptor that mediates the actions of parathyroid hormone (PTH) in the regulation of blood calcium levels, as well as PTH-related protein (PTHrP) in the regulation of skeletal development. Severe loss-of-function homozygous mutations in PTH1R are incompatible with life as in Blomstrand's lethal chondrodysplasia, characterized by accelerated growth plate ossification. More recently, homozygous mutations located in the transmembrane helices, extracellular domains and C-tail of the PTH1R were identified in patients with milder conditions characterized by variable degrees of skeletal and mineral abnormalities.

Background: Eruption sequestrum (ES) is an uncommon condition characterized by irregular calcified tissues on the occlusal surfaces of erupting permanent molars, primarily in children. Despite its rarity, ES has been linked to potential complications, including gingivitis, occlusal trauma, and delayed tooth eruption.

Case Presentation: This paper reports a multiquadrant ES case affecting the first molars of bilateral maxillary and mandibular arches in a 6-year-old pediatric patient.

Cementocytes reside in the cellular cementum of the apical tooth root and resemble bone osteocytes in their markers, lacunocanalicular network, and response to mineralization defects. However, it is unclear if cementocytes have a role in regulating cellular cementum similar to that of osteocytes in controlling bone formation and resorption. The Dmp1Cre-iDTR (Dmp1-DTR) mouse sensitizes -expressing cells, including osteocytes and cementocytes, to diphtheria toxin (DT), allowing selective ablation of cell populations.
