Motivation: DNA methylation-based predictors of various biological metrics have been widely published and are becoming valuable tools in epidemiologic studies of epigenetics and personalized medicine. However, generating these predictors from original source software and web servers is complex and time consuming. Furthermore, different predictors were often derived based on data from different types of arrays, where array differences and batch effects can make predictors difficult to compare across studies.
Results: We integrate these published methods into a single R function to produce 158 previously published predictors for chronological age, biological age, exposures, lifestyle traits and serum protein levels using both classical and principal component-based methods. To mitigate batch and array differences, we also provide a modified RCP method (ref-RCP) that normalize input DNA methylation data to reference data prior to estimation. Evaluations in real datasets show that this approach improves estimate precision and comparability across studies.

In Alzheimer's disease, blood-based DNA methylation signatures are increasingly being explored as minimally invasive peripheral biomarkers. We previously reported associations between blood DNA methylation in the CHI3L1 gene (encoding YKL-40) and cerebrospinal fluid (CSF) levels of YKL-40, a marker of neuroinflammation.

The complexity of epigenetic changes that accompany aging has been distilled into a number of molecular timepieces-termed epigenetic clocks-that characterize the pace of biological aging to differing degrees. Here, we develop and validate a DNA methylation-based Physiological health Age (PhysAge) score, comprised of eight DNA methylation surrogates to represent multi-system physiology and developed from commonly measured clinical biomarkers: CRP, peak flow, pulse pressure, HDL-cholesterol, Hba1c, waist-to-height ratio (WHR), cystatin C, and dehydroepianrosterone sulphate (DHEAS). We use data from the population-representative US Health and Retirement Study (HRS), split into a training (n = 1589) and test sample (n = 1588) and corroborate findings in two independent cohorts: The Irish Longitudinal Study of Aging (TILDA; n = 488) and the Northern Ireland Cohort for the Longitudinal Study of Ageing (NICOLA; n = 1830).

Endometrial cancer (EC) is the most common gynecologic malignancy in developed countries, with a growing incidence and significant molecular heterogeneity that challenges traditional diagnostic and management paradigms. While histopathological assessment remains the gold standard for diagnosis, emerging liquid biopsy technologies provide promising non-invasive alternatives for tumor detection, molecular profiling, and disease monitoring.
