In this procedure we have included an open-source method for a customized operant chamber optimized for long-term miniature microscope (miniscope) recordings. •The miniscope box is designed to function with custom or typical med-associates style accessories (e.g., houselights, levers, etc.).•The majority of parts can be directly purchased which minimizes the need for skilled and time-consuming labor.•We include designs and estimated pricing for a single box but it is recommended to build these in larger batches to efficiently utilize bulk ordering of certain components.

Global efforts to standardise methodologies benefit greatly from open-source procedures that enable the generation of comparable data. Here, we present a modular, high-throughput nucleic acid extraction protocol standardised within the Earth Hologenome Initiative to generate both genomic and microbial metagenomic data from faecal samples of vertebrates. The procedure enables the purification of either RNA and DNA in separate fractions (DREX1) or as total nucleic acids (DREX2).

Antimicrobial peptides (AMPs) have emerged as promising candidates for combating drug-resistant pathogens and certain cancer types. However, their therapeutic applications are often limited by undesired hemolytic activity, while many AMPs exhibit only moderate potency. Herein, the "helical wheel rotation" strategy as a simple, cost-effective, and modular approach to optimize the pharmacological properties of amphipathic α-helical AMPs without altering their amino acid composition is explored.

The direct introduction of a bromine atom into organic molecules is valuable because of its versatility in synthetic intermediates and modular building blocks but traditionally suffers from poor selectivity and relatively complicated and/or harsh reaction conditions. We herein present the first visible-light-driven organic-dye-catalyzed bromination protocol under mild conditions with high regioselectivity. This methodology leverages rapid intramolecular radical trapping to achieve regioselective monobromination of alkenes, thus effectively suppressing competing dibromination and electrophilic bromination pathways.

Background: Multi-epitope vaccines have become the preferred strategy for protection against infectious diseases by integrating multiple MHC-restricted T-cell and B-cell epitopes that elicit both humoral and cellular immune responses against pathogens. Computational methods address various aspects independently, yet their orchestration is technically challenging, as most bioinformatics tools are accessible through heterogeneous interfaces and lack interoperability features. The present work proposes a novel framework for rationalized multi-epitope vaccine design that streamlines end-to-end analyses through an integrated web-based environment.

This editorial examines the feasibility and ethical implications of integrating robotic-assisted orthopaedic surgery into South Africa's healthcare system. While such technologies are well-established in high-income settings, enhancing surgical precision, reproducibility and postoperative outcomes, their widespread adoption in resource-limited contexts poses considerable logistical, financial and ethical challenges. The South African health landscape, marked by stark disparities between public and private sectors, provides a unique setting in which to assess whether such innovations should be prioritised over more pressing service delivery needs.
