CRISPR-Cas are adaptive immune systems in bacteria and archaea that utilize CRISPR RNA-guided surveillance complexes to target complementary RNA or DNA for destruction. Target RNA cleavage at regular intervals is characteristic of type III effector complexes. Here, we determine the structures of the Synechocystis type III-Dv complex, an apparent evolutionary intermediate from multi-protein to single-protein type III effectors, in pre- and post-cleavage states. The structures show how multi-subunit fusion proteins in the effector are tethered together in an unusual arrangement to assemble into an active and programmable RNA endonuclease and how the effector utilizes a distinct mechanism for target RNA seeding from other type III effectors. Using structural, biochemical, and quantum/classical molecular dynamics simulation, we study the structure and dynamics of the three catalytic sites, where a 2'-OH of the ribose on the target RNA acts as a nucleophile for in line self-cleavage of the upstream scissile phosphate. Strikingly, the arrangement at the catalytic residues of most type III complexes resembles the active site of ribozymes, including the hammerhead, pistol, and Varkud satellite ribozymes. Our work provides detailed molecular insight into the mechanisms of RNA targeting and cleavage by an important intermediate in the evolution of type III effector complexes.

Objective: Exploring the clinical effects of the method using elevating the suborbicularis oculi fat (SOOF) and suturing it with the orbital septum and orbital fat as a whole for fixation to correct lower eyelid bags and tear trough deformities.
Methods: A retrospective analysis was conducted on clinical data of patients who underwent transcutaneous lower eyelid blepharoplasty at Hunan Provincial People's Hospital from January 2019 to January 2021. The method involved elevating the suborbicularis oculi fat and suturing it with the orbital septum and orbital fat as a whole for fixation to correct lower eyelid bags and tear trough deformities, and follow-up to evaluate the correction of eyelid bags, tear troughs, and mid-face ptosis, as well as patient satisfaction.

Half-sandwich complexes of platinum-group metals are a widely studied subgroup of organometallic compounds with promising anticancer and antimicrobial properties. Recently, we have published a set of polyhapto arene/arenyl Ru(II), Os(II), Ir(III) and Rh(III) complexes with hetaryl-substituted 1-N-glucopyranosyl-1,2,3-triazole and C-glycopyranosyl-1,3,4- and -1,2,4-oxadiazole-type N,N-bidentate ligands, several of which exhibited (sub)micromolar antineoplastic and bacteriostatic potencies. The structure-activity relationships of these series indicated that the nature of the azole ring and its way of connection to the pyranoid sugar unit played crucial roles in the biological activity of such complexes.

Based on the first-principles calculations, we theoretically investigate the electronic structure, interfacial and optical properties of the tellurene/ZnSe (namely α- and γ-Te/ZnSe) van der Waals heterostructures (vdWHs). In the most stable stacking pattern, the α-Te/ZnSe vdWH exhibits an indirect band gap of 0.41 eV and forms a type-I band alignment, while the γ-Te/ZnSe vdWH possesses a p-type Schottky contact with a favorable Schottky barrier height of 0.

Background: The PORTEC-3 trial investigated the benefit of chemoradiotherapy versus pelvic radiotherapy alone for women with high-risk endometrial cancer. We present the preplanned long-term analysis of the randomised PORTEC-3 trial with a post-hoc analysis including molecular classification of the tumours.
Methods: PORTEC-3 was an open-label, multicentre, randomised, international phase 3 trial.

Respiratory syncytial virus (RSV), the most common cause of bronchiolitis and pneumonia in infants, elicits a remarkably weak innate immune response. This is partly due to type I interferon (IFN) antagonism by the non-structural RSV NS1 protein. It was recently suggested that NS1 could modulate host transcription via an interaction with the MED25 subunit of the Mediator complex.
