Background: The GPNMB single-nucleotide polymorphism rs199347 and GBA1 variants both associate with Lewy body disorder (LBD) risk. GPNMB encodes glycoprotein nonmetastatic melanoma protein B (GPNMB), a biomarker for GBA1-associated Gaucher's disease.
Objective: The aim of this study was to determine whether GPNMB levels (1) differ in LBD with and without GBA1 variants and (2) associate with rs199347 genotype.
Methods: We quantified GPNMB levels in plasma and cerebrospinal fluid (CSF) from 124 individuals with LBD with one GBA1 variant (121 plasma, 14 CSF), 631 individuals with LBD without GBA1 variants (626 plasma, 41 CSF), 9 neurologically normal individuals with one GBA1 variant (plasma), and 2 individuals with two GBA1 variants (plasma). We tested for associations between GPNMB levels and rs199347 or GBA1 status.
Results: GPNMB levels associate with rs199347 genotype in plasma (P = 0.022) and CSF (P = 0.007), but not with GBA1 status.
Conclusions: rs199347 is a protein quantitative trait locus for GPNMB. GPNMB levels are unaltered in individuals carrying one GBA1 variant. © 2024 The Authors. Movement Disorders published by Wiley Periodicals LLC on behalf of International Parkinson and Movement Disorder Society.

ObjectiveTo investigate the role and mechanism of long noncoding RNA LINKA (LncRNA LINKA) in hyperoxia-induced acute lung injury (HALI), specifically focusing on its impact on the GPNMB (glycoprotein nonmetastatic B protein)/HIF-1α (hypoxia-inducible factor 1-alpha) signaling pathway of apoptosis.MethodsAn experimental animal study was conducted using specific pathogen-free (SPF) male C57BL/6 mice and GPNMB knockout (KO) mice. Lung injury was assessed by measuring total protein in bronchoalveolar lavage fluid (BALF), lung wet/dry weight (W/D) ratio, serum levels of inflammatory (tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β)) and oxidative stress (malondialdehyde (MDA) and superoxide dismutase (SOD)) mediators, histopathological scoring (hematoxylin and eosin staining), apoptosis rate (terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL) assay), and expression levels of GPNMB/HIF-1α pathway proteins (p-GPNMB, phosphorylated leucine-rich repeat kinase 2 (p-LRRK2), p-HIF-1α) and apoptosis regulators (BCL2-associated X protein (Bax), B-cell lymphoma 2 (Bcl-2)) via western blotting.

Ischemic stroke (IS) is a complex neurological disorder caused by reduced cerebral blood flow, typically resulting in tissue damage due to hypoxia and nutrient deficiency. Objective: Because white adipose tissue is infiltrated by several immune cells and their signature in individuals with obesity has not been fully characterized, we wanted to study the most abundant population, which is macrophages, a subtype of myeloid cell.
Methods: To address this objective, we performed transcriptomic analysis of subcutaneous adipose tissue (SAT)- and visceral adipose tissue (VAT)-infiltrated CD11b myeloid cells from individuals with severe obesity.
Results: Our results showed that gene expression in human white adipose tissue-infiltrated CD11b myeloid cells was depot-dependent.
