Article Abstract

The aim of the work presented in this manuscript was to radiolabel methotrexate and prepare radiolabeled methotrexate micelles, an antifolate drug with Tc-99m using QbD approach. The radiolabeling was executed using the experimental design and the radiolabeled drug was further encapsulated in micelles. The authors are of the view that the radiolabeled MTX could be used to target the folate receptor overexpressing cancers such as the kidney, colorectal, breast, brain etc thereby opening newer possibilities to the theranostic applications of the formed conjugate.

Significant advances in novel drug delivery systems have led to the development of ligand-conjugated nanoparticles, enabling targeted delivery of therapeutic agents to disease-specific sites. A widely adopted approach for treating rheumatoid arthritis involves targeting folate receptors, which are overexpressed in inflamed tissues. This study focused on formulating methotrexate-loaded, folate-conjugated core-shell polymeric nanoparticles (PF/CS).

Stimuli-responsive polymer-based nanocarriers enhance the drug delivery efficiency by enabling targeted release at tumor sites. However, integrating therapeutic and diagnostic functions into a single nanoplatform while maintaining control over both remains a significant challenge. This study presents a stimuli-responsive, multifunctional poly(ethylene oxide)--poly(ε-caprolactone) (PEO--PCL) nanocarrier for combination cancer therapy and bioimaging.

Background: Ovarian cancer is difficult to detect in its early stages, and it has a high potential for invasion and metastasis, along with a high rate of recurrence. These factors contribute to the poor prognosis and reduced survival times for patients with this disease. The effectiveness of conventional chemoradiotherapy remains limited.

Department of Molecular Biology and Genetics, Faculty of Arts and Science, Necmettin Erbakan University, Konya 42090, Turkey.

In the present study, ultra-small, magnetic, oleyl amine-coated FeO nanoparticles were synthesized and stabilized with a cationic ligand, cetyltrimethylammonium bromide, and an anticancer drug, methotrexate, was incorporated into a micelle-like nanoparticle structure for glioblastoma treatment. Nanoparticles were further characterized for their physicochemical properties using spectroscopic methods. Drug incorporation efficiency, drug loading, and drug release profile of the nanoparticles were investigated.

Ovarian cancer, a highly lethal form of gynecological cancer globally, has witnessed notable advancements in its treatment through the integration of nanotechnology and immunotherapy. Here, we designed a novel astragalus polysaccharide vector (PDA), encapsulating podophyllotoxin (PPT), and modifying methotrexate (DSPE-PEG-MTX) on its surface for targeting ovarian cancer cells with high folate receptor expression. We prepared novel MTX-modified PPT-loaded astragalus polysaccharide micelles (MTX-PPT-micelles) by dialysis method and evaluated their characterization, stability, safety and targeting ability.
