Article Abstract

. Severe radiation-induced lymphopenia occurs in 40% of patients treated for primary brain tumors and is an independent risk factor of poor survival outcomes. We developed anframework that estimates the radiation doses received by lymphocytes during volumetric modulated arc therapy brain irradiation.. We implemented a simulation consisting of two interconnected compartmental models describing the slow recirculation of lymphocytes between lymphoid organs (M1) and the bloodstream (M2). We used dosimetry data from 33 patients treated with chemo-radiation for glioblastoma to compare three cases of the model, corresponding to different physical and biological scenarios: (H1) lymphocytes circulation only in the bloodstream i.e. circulation inM2only; (H2) lymphocytes recirculation between lymphoid organs i.e. circulation inM1andM2interconnected; (H3) lymphocytes recirculation between lymphoid organs and deep-learning computed out-of-field (OOF) dose to head and neck (H&N) lymphoid structures. A sensitivity analysis of the model's parameters was also performed.. For H1, H2 and H3 cases respectively, the irradiated fraction of lymphocytes was 99.8 ± 0.7%, 40.4 ± 10.2% et 97.6 ± 2.5%, and the average dose to irradiated pool was 309.9 ± 74.7 mGy, 52.6 ± 21.1 mGy and 265.6 ± 48.5 mGy. The recirculation process considered in the H2 case implied that irradiated lymphocytes were irradiated in the field only 1.58 ± 0.91 times on average after treatment. The OOF irradiation of H&N lymphoid structures considered in H3 was an important contribution to lymphocytes dose. In all cases, the estimated doses are low compared with lymphocytes radiosensitivity, and other mechanisms could explain high prevalence of RIL in patients with brain tumors.. Our framework is the first to take into account OOF doses and recirculation in lymphocyte dose assessment during brain irradiation. Our results demonstrate the need to clarify the indirect effects of irradiation on lymphopenia, in order to potentiate the combination of radio-immunotherapy or the abscopal effect.

Abnormal expression of HLA class Ib, MICA and MICB molecules is associated with the evolution of pathological conditions and clinical settings. Here, we use RNA-sequencing data from two publicly-available projects, from different human organs and tissues and at single-cell level, to present their transcriptional expression throughout the human body, in comparison to that of HLA class Ia, HLA class II, their costimulatory molecules, and the main HLA transcription factors. Our analyses for 21 target genes reveal that median gene expression differs by orders of magnitude and that the classical/non-classical HLA distinction is not absolute for overall expression.

Unlabelled: B-cell lymphomas are highly aggressive forms of lymphoma that commonly present with lymphadenopathy, systemic "B" symptoms, or organ involvement making them easy to recognize; however, a small percentage of B-cell lymphomas can present without any typical symptoms or evidence of lymphadenopathy, resulting in delayed recognition and management. Isolated thrombocytopenia without anaemia or leukopenia is an unusual presentation of B cell lymphomas and may be misdiagnosed as immune thrombocytopenia (ITP). Given the rarity of this presentation, we wish to report a case of a 76-year-old female who presented with palpitations, shortness of breath, and recurrent chest infections.

Advances in the immunometabolism field have shown that infiltrated immune cells play a pivotal role in the development and function of thermogenic adipose tissue (TAT), including brown and beige fat. However, scarce research has focused on the role that organized lymphoid structures, like lymph nodes and lymphatics vessels, may exert on TAT. In this review we summarize the evidence suggesting that a significant link exists between the lymphoid tissues and adipose tissue, and we describe the most important in vitro and in vivo findings indicating that organized lymphoid tissues also play an important role in TAT biogenesis and function, raising relevant questions which are still unsolved in this emerging field.

B cells are critical components of the adaptive immune system that proliferate and differentiate within the secondary lymphoid organs upon recognition of antigens and engagement of T cells. Traditional two-dimensional (2D) cell cultures fall short of replicating the intricate structures and dynamic evolution of three-dimensional (3D) environments found in lymphoid organs, prompting the development of more physiologically pertinent models. Our approach employs -hexanoyl glycol chitosan (HGC) coated ultra-low attachment (ULA) lattice plates to cultivate a 3D co-culture of CD40L-expressing MS5 stromal cells and naïve B cells derived from the peripheral blood mononuclear cells (PBMCs) of healthy human donors.
