Introduction: Childhood acute lymphoblastic leukemia (cALL), the most common pediatric hematologic malignancy, arises primarily from B-cell origin and is strongly associated with immune dysfunction. This article integrated single-cell and bulk transcriptomic data to identify key B-cell subsets and cALL-related molecules as biomarkers.
Methods: Single-cell RNA sequencing (scRNA-seq) Data from 2 pre-B high hyperdiploid (HHD) ALL patients and 3 healthy pediatric bone marrow samples (GSE132509) were utilized for cell clustering using the Seurat package.

The IKAROS (IKZF1) transcription factor regulates the differentiation of pre-B cells to mature B lymphocytes. The loss of IKZF1 expression has been linked with increased matrix adhesion along with increased expression of stemness markers that form a hallmark of B-lymphoblasts. However, the spectrum of differentially regulated genes has not been investigated in detail in B-cell acute lymphoblastic leukaemia (B-ALL) patients expressing IKZF1 deletion.

A previously healthy five-year-old boy was seen in the rheumatology clinic with a five-month history of migrating joint discomfort, followed by right knee and left ankle arthritis. He showed no signs of lymphadenopathy, organomegaly, rashes, or fever. He could not bear weight and had nocturnal pain that was out of proportion to juvenile idiopathic arthritis.

Background: L-asparaginase is an essential chemotherapeutic agent for treating pediatric acute lymphoblastic leukemia (ALL), but its use is commonly linked to severe complications, including pancreatitis. Pancreatic pseudocysts and panniculitis are a rare complication of L-asparaginase-induced pancreatitis.
Case Presentation: We report the case of a 20-month-old girl diagnosed with pre-B ALL who developed pancreatitis, complicated by pseudocyst formation and panniculitis, following L-asparaginase therapy.
