Heterozygosity of () class I genes is linked to beneficial outcomes after HIV infection, presumably through greater breadth of HIV epitope presentation and cytotoxic T cell response. Distinct allotype pairs, however, differ in the extent to which they bind shared sets of peptides. We developed a functional divergence metric that measures pairwise complementarity of allotype-associated peptide binding profiles. Greater functional divergence for pairs of HLA-A and/or HLA-B allotypes was associated with slower AIDS progression and independently with enhanced viral load control. The metric predicts immune breadth at the peptide level rather than gene level and redefines heterozygosity as a continuum differentially affecting disease outcome. Functional divergence may affect response to additional infections, vaccination, immunotherapy, and other diseases where heterozygote advantage occurs.

Tandem repetition is one of the major processes underlying genome evolution and phenotypic diversification. While newly formed tandem repeats are often easy to identify, it is more challenging to detect repeat copies as they diverge over evolutionary timescales. Diversity-oriented synthesis (DOS) has emerged as an efficient strategy for constructing diverse compound libraries, facilitating hit or lead identification in the drug discovery process. The big-headed turtle ( ), currently the only extant member of the genus and the family Platysternidae, has undergone severe population declines driven by poaching, illegal trade, and habitat loss, leading to its classification as Critically Endangered (CR) by the International Union for Conservation of Nature (IUCN). Despite its conservation status, persistent taxonomic ambiguities and unresolved phylogenetic relationships have hindered effective protection and management. This study integrated evidence from genome-wide single nucleotide polymorphisms (SNPs), mitochondrial DNA sequences ( , ), and morphological data to reconstruct the phylogeny and phylogeography of and revise its taxonomy.
