Autologous CD19-targeting CAR-T has transformed management of relapsed/refractory adult B-cell acute lymphoblastic leukaemia(B-ALL) but relapse post-allogeneic stem cell transplant(allo-SCT) is frequently accompanied by profound lymphopenia, impaired T-cell fitness and aggressive disease requiring urgent treatment, making autologous CAR-T challenging to deliver. We developed an allogeneic matched-donor CD19CAR product (CAR-DLI) for adult B-ALL following allo-SCT failure. Here we evaluate the risks/benefits of pre-CAR-DLI lymphodepleting chemotherapy (LD), and the efficacy of repeat CAR-DLI dosing as per conventional DLI scheduling/protocols.

Canine distemper virus (CDV) and mink enteritis virus (MEV) are among the most devastating viruses in minks (Mustela vison). They often cause two fatal diseases-canine distemper (CD) and mink viral enteritis (MVE)-resulting in severe respiratory symptoms and acute enteritis in breeding minks, respectively. Here, using the attenuated CDV vaccine strain CDV3-CL (currently employed in breeding mink in China) as a backbone, we constructed a recombinant strain, rCDV3-mVP2, expressing the wild-type MEV capsid protein VP2.

The IFN-type-I pathway is involved in radiotherapy (RT)-mediated immune responses. Large RT fractions have been suggested to potently induce this pathway. Neoadjuvant hypofractionated short-course (scRT) and conventional long-course (lcRT) RT applied for the treatment of locally advanced rectal adenocarcinoma patients provides a unique model to address the immuno-stimulatory properties of RT on a systemic level.
