Recent evidence indicates that the concentration of ATP remains stable during neuronal activity due to activity-dependent ATP production. However, the mechanisms of activity-dependent ATP production remain controversial. To stabilize the ATP concentration, feedforward mechanisms, which may rely on calcium or the sodium-potassium pump, do not require changes in the ATP and ADP concentrations.

Drug delivery to the central nervous system (CNS) is primarily hindered by the blood-brain barrier (BBB). Glioblastoma (GB), IDH-wildtype (IDH-wt), is the most prevalent primary malignant brain neoplasm in adults. Despite adjuvant therapy, the prognosis for these tumors remains dismal, with a median survival of around 15-18 months. Although rare, extracranial metastases from GB are reported with increasing frequency, likely due to advancements in follow-up, treatments, and improved patient survival.
