Assessment of drug-induced effects on the cardiovascular (CV) system remains a critical component of the drug discovery process enabling refinement of the therapeutic index. Predicting potential drug-related unintended CV effects in the preclinical stage is necessary for first-in-human dose selection and preclusion of adverse CV effects in the clinical stage. According to the current guidelines for small molecules, nonclinical CV safety assessment conducted via telemetry analyses should be included in the safety pharmacology core battery studies. However, the manual for quantitative evaluation of the CV safety signals in animals is available only for electrocardiogram parameters (i.e., QT interval assessment), not for hemodynamic parameters (i.e., heart rate, blood pressure, etc.). Various model-based approaches, including empirical pharmacokinetic-toxicodynamic analyses and systems pharmacology modeling could be used in the framework of telemetry data evaluation. In this tutorial, we provide a comprehensive workflow for the analysis of nonclinical CV safety on hemodynamic parameters with a sequential approach, highlight the challenges associated with the data, and propose respective solutions, complemented with a reproducible example. The work is aimed at helping researchers conduct model-based analyses of the CV safety in animals with subsequent translation of the effect to humans seamlessly and efficiently.

Trauma-informed communication has shown promise in healthcare settings for supporting individuals affected by adverse childhood experiences (ACEs), yet its application in public health messaging remains underexplored. Given the strong link between ACEs and intimate partner violence victimization (IPVV), this study designed and tested a trauma-informed, text-based message aimed at promoting trauma understanding and positive behavioral intentions among women experiencing IPVV with self-reported ACEs. The message incorporated two core trauma-informed components-psychoeducation and empowerment-and was evaluated against a conventional IPVV message in a randomized controlled trial ( = 289).

The single nucleotide polymorphism, rs738409, is the strongest known genetic risk factor for metabolic dysfunction-associated steatotic liver disease; thus, targeting the minor allele with a GalNAc-conjugated siRNA is an attractive strategy to treat patients carrying the genetic variant. To enable translational safety assessment of a GalNAc-conjugated siRNA that specifically targets the rs738409 sequence of , a transgenic human knock-in mouse (hu) was utilized. This model showed no significant genotype-related phenotypic differences to wild-type mice in a phenotype characterization study when maintained on standard rodent chow.

High-altitude electrical work is a high-risk and physically demanding occupation that has received limited empirical investigation, particularly regarding the physiological and psychological fatigue experienced during operations. This study provides an initial empirical exploration into fatigue among high-altitude electrical workers by examining the relationship between subjective fatigue perception and multiple physiological indicators in realistic working environments. Thirty professional high-altitude electrical workers participated in field-based measurements conducted during routine summer operations.

Type 2 (T2) inflammation underlies a substantial subset of moderate-to-severe asthma, contributing to persistent symptoms and frequent exacerbations. Dupilumab, a fully human immunoglobulin G subclass 4 (IgG4) monoclonal antibody, targets the interleukin-4 receptor alpha (IL-4Rα), thereby inhibiting both interleukin-4 (IL-4) and interleukin-13 (IL-13) signaling-which are key cytokines driving T2 inflammation. This review examines the formulation, pharmacological profile, clinical efficacy, safety, and cost-effectiveness of dupilumab in the treatment of asthma, with an emphasis on its role across T2-high and selected T2-low phenotypes.
