Article Abstract

Novel nickel(II) and copper(II) complexes of 2-(,-dimethylformamidine)-3-formyl-5,10,15,20-tetraarylporphyrins have been synthesized for the first time from 2-aminoporphyrins under Vilsmeier-Haack conditions. These porphyrins are utilized as new building blocks to construct diverse β-pyrimidine-fused 5,10,15,20-tetraarylporphyrins in good yields a cascade ammonia-mediated condensation and intramolecular aza-6π-annulation/aromatization in 1,2-dichloroethane at 80 °C. Furthermore, copper(II) β-pyrimidine-fused porphyrins underwent demetallation in the presence of conc. HSO to afford free-base porphyrins, which on zinc insertion using Zn(OAc) in CHCl-MeOH provided zinc(II) β-pyrimidine-fused porphyrins in appreciable yields. Notably, these newly synthesized π-extended porphyrins displayed a modest bathochromic shift in their electronic absorption and emission spectra as compared to the traditional -tetraarylporphyrins. However, the protonated porphyrins () and () displayed a significant red-shifted absorption.

Yes-associated protein (YAP) is a major downstream nuclear coactivator of the Hippo pathway and is activated during myocardial hypertrophy. Verteporfin, a YAP inhibitor, may serve as a potential treatment for myocardial hypertrophy. This study was aimed at exploring the role and underlying mechanisms of verteporfin in isoproterenol (ISO)-induced myocardial hypertrophy both in vivo and in vitro.

Gold/Prussian Blue-Based Nanocomposites with Dual Nanozyme Activities Exert a Synergistic Effect of Starvation Therapy and Sonodynamic Therapy in the Treatment of Liver Cancer.

School of Pharmaceutical Sciences, Key Laboratory of Targeting Therapy and Diagnosis for Critical Diseases, Zhengzhou University, Zhengzhou, 450001, People's Republic of China.

Purpose: This study aimed to develop a composite nanozyme system (Au/PB-Ce6-HA) based on gold nanoparticles (AuNPs) and Prussian blue nanoparticles (PBNPs) to combat tumor hypoxia and insufficient endogenous hydrogen peroxide (HO) deficiency, thus enhancing the efficacy of sonodynamic therapy (SDT) and starvation therapy for liver cancer.

Methods: The Au/PB-Ce6-HA system was constructed by in situ embedding AuNPs on PBNPs, loading the sonosensitizer Chlorin e6 (Ce6), and surface-coating with thiolated hyaluronic acid (HA-SH). The system was evaluated both in vitro and in vivo to assess its ability to catalyze glucose to generate HO, decompose HO to produce oxygen, and generate highly toxic reactive oxygen species (ROS) under ultrasound irradiation.

The title complex, [Fe(CHN)(CHN)]·3CH, possesses inversion symmetry with the iron(II) atom located on a center of symmetry. The metal atom is coordinated in a symmetric octa-hedral geometry by four pyrrole N atoms of the porphyrin ligand in the equatorial plane and two N atoms of 1-methyl-imidazole ligands in the axial sites; the complex crystallizes with three toluene solvent mol-ecules. The average Fe-N (N is a porphyrin N atom) bond length is 1.

Background: Cisplatin (DDP) is a clinical first-line chemotherapy drug for hepatocellular carcinoma (HCC), but treatment is often ineffective due to drug resistance. Yes-associated protein 1 (YAP1) is a critical regulator/factor in HCC tumor progression. Our previous research showed that DDP promoted the expression of YAP1 in mice bearing H22 cell in situ liver tumors, which might be related to the poor therapeutic effect of DDP.

Phosphorus(V)-centered porphyrins (P(V)-porphyrins) are an important class of functional dyes in many fields of research, and axial ligands on the phosphorus atom affect the electronic properties of P(V)-porphyrins and add functions. Herein, we report on the synthesis and characterization of a hitherto unknown P(V)-porphyrin having hydrogen atoms as axial ligands (1·PF , PF is a counter anion). Synthesis of 1·PF was achieved by treatment of dichloro-derivative (2·Cl) with LiAlH followed by AgPF via hydride reduction accompanied by one-electron reduction and one-electron oxidation.
