Recognizing animal emotions is critical to their welfare and can lead to a better relationship with humans and the environment, especially in a widespread species like the donkey, which is often prone to welfare issues. This study aims to assess the emotional response of donkeys through an operant conditioning task with two presumed different emotional contents. Specifically, a within-subject design including positive and negative conditions was conducted, collecting behavioural and physiological (heart rate variability and HRV) parameters. Facial expressions, postures, and movements were analysed by principal component analysis and behavioural diversity indexes (frequencies, activity budgets, richness, Shannon and Gini-Simpson). During the positive condition, both ears were held high and sideways (left: r = -0.793, < 0.0001; right: r = -0.585, = 0.011), while the ears were frontally erected (left: r = 0.924, < 0.0001; right: r = 0.946, < 0.0001) during the negative one. The latter was also associated with an increased tendency to walk (r = 0.709, = 0.001), walk away (r = 0.578, = 0.012), more frequent changes in the body position (V = 0, = 0.022), and greater behavioural complexity (V = 4, = 0.027). As for HRV analysis, the root mean square of successive beat-to-beat differences (rMSSD) was significantly lower after the negative condition. These non-invasive parameters could be considered as possible indicators of donkeys' emotional state.

This study aimed to identify brain activity modulations associated with different types of visual tracking using advanced functional magnetic resonance imaging techniques developed by the Human Connectome Project (HCP) consortium. Magnetic resonance imaging data were collected from 27 healthy volunteers using a 3-T scanner. Statistical Parametric Mapping (SPM) has been profoundly influential to neuroimaging as it has fostered rigorous, statistically grounded structure for model-based inferences that have led to mechanistic insights about the human brain over the past 30 years. The statistical constructs shared with the world through SPM have been instrumental for deriving meaning from neuroimaging data; however, they require simplifying assumptions which can provide results that, while statistically sound, may not accurately reflect the mechanisms of brain function. A platform that fosters the exploration of the rich and varying neuronal and physiologic underpinnings of the measured signals and their associations to behavior and physiologic measures needs a different set of tools.

Vascularization of implanted biomaterials is critical to reconstructive surgery and tissue engineering. Ultimately, the goal is to promote a rapidly perfusable hierarchical microvasculature that persists with time and can meet underlying tissue needs. We have previously shown that using a microsurgical technique, termed micropuncture (MP), in combination with porous granular hydrogel scaffolds (GHS) fabricated via interlinking hydrogel microparticles (microgels) results in a rapidly perfusable patterned microvasculature.
