The reaction of mercaptoacetic acid esters with pentachloro-2-nitro-1,3-butadiene provides the appropriate precursors for the synthesis of 2,3,4-trisubstituted benzo[]quinolines. These heterocycles are easily accessible via a single-step reaction with naphthalen-1-amine or anthracen-1-amine as the precursor. Due to the steric bulk and high electron density ring, the ring closure of benzo[]quinolines takes place exclusively. Such highly substituted annelated pyridine systems can be modified in subsequent, selective reactions to build up new -heterocycles with promising microbiological properties. The antibacterial and antiproliferative assays against four mammalian cell lines demonstrate that some of the sulfur-substituted benzo[]quinoline analogs display potent phenotypic bioactivities in the single-digit micromolar range.

In this paper, we investigated the sensing mechanism of 3-hydroxy-2-(4-(pyrrolidin-1-yl) phenyl) benzo[] quinolin-4(1)-one (PBQ) for detecting phosgene (COCl), employing DFT and TD-DFT. Computational results showed that PBQ underwent the coupled process of TICT and ESIPT. It was further explained that the experimentally observed dual fluorescence phenomenon originated from the co-contribution of the enol* and keto* forms.

This study unveils the first rhodium-catalyzed synthesis of benzo[,]quinoline-spiro-succinimides C-H activation/annulation of 1-naphthylamides with maleimides, utilizing picolinamide as a directing group. This method boasts a broad substrate scope, excellent yields, and wide range of functional group tolerance. Mechanistic insights and promising photoluminescence are reported, demonstrating an efficient route to valuable all-carbon quaternary spirocyclic compounds.
