Article Abstract

Background: Charcot-Marie-Tooth disease (CMT) is a genetically and clinically heterogeneous group of inherited neuropathies. Monoallelic pathogenic variants in ATP1A1 were associated with axonal and intermediate CMT. ATP1A1 encodes for the catalytic α1 subunit of the Na/ K ATPase. Besides neuropathy, other associated phenotypes are spastic paraplegia, intellectual disability, and renal hypomagnesemia. We hereby report the first demyelinating CMT case due to a novel ATP1A1 variant.

Methods: Whole-exome sequencing on the patient's genomic DNA and Sanger sequencing to validate and confirm the segregation of the identified p.P600R ATP1A1 variation were performed. To evaluate functional effects, blood-derived mRNA and protein levels of ATP1A1 and the auxiliary β1 subunit encoded by ATP1B1 were investigated. The ouabain-survival assay was performed in transfected HEK cells to assess cell viability, and two-electrode voltage clamp studies were performed in Xenopus oocytes.

Results: The variant was absent in the local and global control datasets, falls within a highly conserved protein position, and is in a missense-constrained region. The expression levels of ATP1A1 and ATP1B1 were significantly reduced in the patient compared to healthy controls. Electrophysiology indicated that ATP1A1 injected Xenopus oocytes have reduced Na/ K ATPase function. Moreover, HEK cells transfected with a construct encoding ATP1A1 harbouring variants that confers ouabain insensitivity displayed a significant decrease in cell viability after ouabain treatment compared to the wild type, further supporting the pathogenicity of this variant.

Conclusion: Our results further confirm the causative role of ATP1A1 in peripheral neuropathy and broaden the mutational and phenotypic spectrum of ATP1A1-associated CMT.

Introduction: Type 2 diabetes (T2D) is characterized by insulin resistance and chronic inflammation, with macrophages playing a crucial role in pancreatic islet dysfunction. This study explored the intersection of macrophage-specific gene expression and abnormal blood monovalent inorganic cation concentration-related genes (ABRGs) in T2D patients via single-cell RNA sequencing (scRNA-seq) and machine learning to identify key genes and potential therapeutic targets.

Methods: ScRNA-seq data from the pancreatic islet cells of 27 nondiabetic (ND) patients and 17 T2D patients were analyzed to identify differentially expressed genes (DEGs) in macrophages.

Safety of a novel dual-chamber vial for corneal preservation.

Exp Eye Res

August 2025

Department of Ophthalmology, Bascom Palmer Eye Institute, University of Miami Miller School of Medicine, Miami, FL, USA. Electronic address:

Our group developed a novel dual-chamber corneal storage vial (DCV) comprised of two compartments that become isolated by the corneal button and prevent epithelial and endothelial interaction. We hypothesize that the DCV results in similar levels of corneal swelling and corneal endothelial cell (EC) death compared to the standard vial (SCV) and is safe for corneal storage. To assess corneal thickness and EC density, human cornea pairs (N = 18) were recovered by the Florida Lions Eye Bank, with one cornea from each pair stored in the SCV and the other in the DCV.

Degeneration of the stria vascularis in quiet-aged gerbils: Linking structural, cellular and molecular changes to cochlear function.

Presbycusis, or age-related hearing loss, is a prevalent condition characterized by progressive auditory decline, significantly impacting quality of life in older adults. While sensorineural damage has been widely studied, degeneration of the stria vascularis (SV) remains underexplored despite its essential role in cochlear ion homeostasis. The SV is organized into three cellular layers-marginal, intermediate, and basal cells-each with distinct functions critical for maintaining the endocochlear potential.

Background: Clear cell renal cell carcinoma (KIRC) is the most aggressive renal carcinoma subtype of renal carcinoma, characterized by high mortality, early metastasis, and resistance to treatment. Ammonia-induced cell death (AICD) has recently been identified as a novel metabolic mechanism influencing tumor progression, yet its prognostic implication and regulatory networks in KIRC remain underexplored.

Methods: Transcriptomic and clinical information from the TCGA-KIRC cohort and the validation cohort (E-MTAB-1980) were analyzed.
