Highly specialized enamel matrix proteins (EMPs) are predominantly expressed in odontogenic tissues and diverged from common ancestral gene. They are crucial for the maturation of enamel and its extreme complexity in multiple independent lineages. However, divergence of EMPs occured already before the true enamel evolved and their conservancy in toothless species suggests that non-canonical functions are still under natural selection. To elucidate this hypothesis, we carried out an unbiased, comprehensive phenotyping and employed data from the International Mouse Phenotyping Consortium to show functional pleiotropy of amelogenin, ameloblastin, amelotin, and enamelin, genes, i.e. in sensory function, skeletal morphology, cardiovascular function, metabolism, immune system screen, behavior, reproduction, and respiratory function. Mice in all KO mutant lines, i.e. amelogenin KO, ameloblastin KO, amelotin KO, and enamelin KO, as well as mice from the lineage with monomeric form of ameloblastin were affected in multiple physiological systems. Evolutionary conserved motifs and functional pleiotropy support the hypothesis of role of EMPs as general physiological regulators. These findings illustrate how their non-canonical function can still effect the fitness of modern species by an example of influence of amelogenin and ameloblastin on the bone physiology.

Objective: This study aimed to investigate the influence of human amelogenin (AM), amelotin (AMTN), and ameloblastin c-terminal peptide (AMBN-C), both individually and in combination, on the in vitro remineralization of enamel.
Design: AM, AMTN, and AMBN-C were recombined and purified in vitro and formulated into 200 µg/ml chitosan (CS) protein gels. Nine different gel groups, including saline, CS, CS-AM, CS-AMTN, CS-AMBN-C, CS-AM-AMTN, CS-AM-AMBN-C, CS-AMTN-AMBN-C, and CS-AM-AMTN-AMBN-C, were applied to artificial carious tooth samples.

Biomineralization of skeletal and dental tissues has evolved via a suite of regulatory extracellular matrix proteins. The secretory calcium-binding phosphoproteins (SCPPs) are encoded by genes that arose by duplication. In the human genome, 23 SCPP genes have been identified, and 2 groups of SCPPs regulate dental mineralization: bone, dentin, and/or cementum matrix proteins and enamel proteins.

The biological sex estimation of human individuals can be achieved by extracting fragments of the amelogenin protein from small areas of tooth enamel. The amelogenin gene can be found on both sex chromosomes (X and Y) with chromosome-specific differences in its sequence, and consequently the sequences of the expressed protein in teeth. Virtually all current analytical techniques used to identify the occurrence of the male Y chromosome-specific proteoform employ proteoform-specific peptide analysis by LC-ESI MS/MS, which typically results in longer analytical times due to the LC separation step, despite recent efforts of shortening the LC step.
