Article Abstract

Viperin, also known as radical S-adenosyl methionine domain-containing protein (RSAD2) is a multifunctional interferon-stimulated gene (ISG) that is activated during the viral infections. Viperin belongs to S-adenosyl methionine (SAM) superfamily of enzymes known to catalyze radical-mediated reactions and viperin inhibits a wide range of DNA and RNA viruses through its broad range of activity. The present study reports cloning and expression of bovine viperin in a bacterial expression system. PCR-based site-directed mutagenesis was carried out for deletion of N-terminal 1-70 amino acid containing amphipathic helix of viperin that interferes in protein expression and purification. The resultant truncated viperin protein was expressed in , BL-21(DE3) competent cells and purified using nickel charged affinity column. The truncated 54 kDa protein was confirmed by western blot using human RSAD2 as a probe. Further, in house, hyperimmune serum was raised against the truncated viperin in the rabbit and the reactivity was confirmed by western blot using mammalian expression vector construct of viperin transfected in Baby Hamster kidney (BHK) cells and in MDBK cells infected with Foot and Mouth disease Asia I virus.

Insect Janus kinase/signal transducer and activator of transcription (JAK/STAT) pathway plays a crucial role in antiviral defense, yet how arboviruses activate this pathway remains unclear. Here, we reveal that outer capsid protein of a double-stranded RNA (dsRNA) arbovirus (rice gall dwarf virus, RGDV) directly binds the Dome receptor's extracellular domain, triggering receptor dimerization and JAK/STAT activation. Structural and mutational analyses pinpoint cysteine 743 in Dome as essential for viral recognition, establishing Dome as the first identified viral pattern recognition receptor (PRR) in insects capable of initiating JAK/STAT signaling, a pathway previously thought to be exclusively cytokine dependent.

Ubiquitination, an essential post-translational regulatory mechanism, is primarily controlled by E3 ubiquitin ligases that dictate substrate specificity. Following microbial invasion, the innate immune defense mechanism is rapidly activated, wherein transforming growth factor-β-activated kinase 1 (MAP3K7/TAK1), a pivotal node in innate immune signaling, is tightly regulated by E3 ubiquitin ligases that dictate its stability and activity. Among these, Ring Finger Protein 34 (RNF34), a member of the E3 ligase family, has a role in regulating innate immunity in teleost fish that remains uncharacterized.

In mammals, cyclic GMP-AMP synthase (cGAS) senses cytosolic dsDNA to trigger type I interferon (IFN) response via the sting signalling, but its regulatory role in fish remains unclear. In the present study, cGAS and cGAS-like (cGASL) were identified in the snakehead (Channa argus), but they differ in the organization of two domains, NTase domain, MAB21 domain, with an additional transmembrane domain in cGASL. It is further revealed that cGAS can bind with Sting to enhance the production of type I IFN and IFN-stimulated genes (ISGs), such as viperin, mx, and its entire molecule with all domains is necessary for its functioning.
