Alternative use of short distance tandem sites such as NAGN AG are a common mechanism of alternative splicing; however, single nucleotide variants are rarely reported as likely to generate or to disrupt tandem splice sites. We identify a pathogenic intron 5 STK11 variant (NM_000455.4:c.[735-6A>G];[=]) segregating with the mucocutaneous features but not the hamartomatous polyps of Peutz-Jeghers syndrome in two individuals. By RNAseq analysis of peripheral blood mRNA, this variant was shown to generate a novel and preferentially used tandem proximal splice acceptor (AAGTGAAG). The variant transcript (NM_000455.4:c.734_734 + 1insTGAAG), which encodes a frameshift (p.[Tyr246Glufs*43]) constituted 36%-43% of STK11 transcripts suggesting partial escape from nonsense mediated mRNA decay and translation of a truncated protein. A review of the ClinVar database identified other similar variants. We suggest that nucleotide changes creating or disrupting tandem alternative splice sites are a pertinent disease mechanism and require contextualization for clinical reporting. Additionally, we hypothesize that some pathogenic STK11 variants cause an attenuated phenotype.

The Sensomics approach, including activity-guided fractionation and taste dilution analysis, was employed to identify the key compounds responsible for the bitter off-taste of sunflower press cake. A combination of liquid chromatography-tandem mass spectrometry, liquid chromatography-time-of-flight-mass spectrometry, one-/two-dimensional nuclear magnetic resonance spectroscopy, and dose-overthreshold factor calculation led to the identification of 9,12,13-trihydroxyoctadec-10-enoic acid, 9,10,11-trihydroxyoctadec-12-enoic acid, 11,12,13-trihydroxyoctadec-9-enoic acid, (10,12)-9-hydroxyoctadeca-10,12-dienoic acid, (10,12)-9-hydroxyoctadeca-10,12-dienoic acid, (9,11)-13-hydroxyoctadeca-9,11-dienoic acid, (9,11)-13-hydroxyoctadeca-9,11-dienoic acid, (9,11)-13-oxooctadeca-9,11-dienoic acid, α-linolenic acid, linoleic acid, oleic acid, 2-hydroxyoleic acid, palmitic acid, stearic acid, and novel pinocarveol β-d-apiofuranosyl-(1→6)-β-d-(4--caffeoyl) glucopyranoside as contributors to the bitterness of sunflower press cake. The findings provide valuable insights into the sensory challenges associated with using sunflower press cake in food applications and offer pathways to enhance its palatability and potential as a sustainable protein alternative to meet future protein demands.

Objective: The delivery of circulating insulin to skeletal muscle myocytes is a rate-limiting step in peripheral insulin action, and there is minimal understanding of the underlying mechanisms in endothelial cells. Recognizing that the LDL receptor family member apolipoprotein E receptor 2 (ApoER2, also known as LRP8) mediates apolipoprotein E (ApoE)-induced signaling in endothelial cells, the present project determined if endothelial ApoER2 influences glucose homeostasis in mice.
Methods: Mice were generated deficient in ApoER2 selectively in endothelial cells, and glucose homeostasis was studied.

Drug monitoring of antidepressants in plasma and oral fluid represents a valuable tool in clinical practice, enabling the optimisation of treatment efficacy and the reduction of adverse effects. Given the significant interindividual variability in antidepressant response-driven by factors such as metabolism, drug-drug interactions, and adherence to therapy-drug monitoring facilitates dose adjustment based on measured drug concentrations, ensuring levels remain within the therapeutic window. This study aimed at developing and validating a robust, rapid, and sensitive method for the simultaneous quantification of 21 selected antidepressants and their metabolites in only 100 μL of plasma and oral fluid.
