Fluorescent Correlation Spectroscopy (FCS) allows us to determine interactions of labeled proteins or changes in the oligomeric state. The FCS method needs a low amount of fluorescent dye, near nanomolar concentrations. To control the amount of fluorescent dye, we used new photoconvertible FP SAASoti. This work is devoted to the proof of principle of using photoconvertible proteins to measure caspase enzymatic activity in a single live cell. The advantage of this approach is that partial photoconversion of the FP makes FCS measurements possible when studying enzymatic reactions. To investigate the process, in vivo we used HeLa cell line expressing the engineered FRET sensor, SAASoti-23-KFP. This FRET sensor has a cleavable (DEVD) sequence in the linker between two FPs for the detection of one of the key enzymes of apoptosis, caspase-3. Caspase-3 activity was detected by registering the increase in the fluorescent lifetimes of the sensor, whereas the diffusion coefficient of SAASoti decreased. This can be explained by an increase in the total cell viscosity during apoptosis. We can suppose that in the moment of detectible caspase-3 activity, cell structure already has crucial changes in viscosity.

Background: Interventions aimed to increase healthcare provider empathy and capacity to deliver person-centered care have been shown to improve healthcare seeking and outcomes. In the context of self-injectable contraception, empathetic counseling and coaching may be promising approaches for addressing "fear of the needle" among clients interested in using subcutaneous depot medroxyprogesterone (DMPA-SC). In Nigeria, the Delivering Innovation in Self-Care (DISC) project developed and evaluated an empathy-based in-service training and supportive supervision intervention for public sector family (FP) planning providers implemented in conjunction with community-based mobilization.

In Tl myocardial perfusion single-photon emission computed tomography (SPECT), gastric wall uptake can impact the inferior wall. This study aimed to evaluate the effectiveness and usefulness of the masking on un-smoothed image (MUS) method for Tl myocardial perfusion SPECT. A hemispherical gastric wall phantom was created to simulate the gastric fundus located closest to the myocardium, and the activity was enclosed to achieve an SPECT count ratio against the myocardium equivalent to that observed in clinical practice.

Type 2 diabetes (T2D) is characterized by persistent and unresolved tissue inflammation caused by the infiltration and dysregulation of immune cells. Current therapeutics targeting inflammatory immune cells for T2D remain limited. In this study, we analyzed single cell RNA from metabolic organs in T2D, revealing increased macrophage accumulation and a pathogenic macrophage subpopulation defined as NOD-like receptor (NLR) family pyrin domain-containing 3 (NLRP3) inflammatory and metabolically activated macrophages.
