Immunoglobulins type-M (IgMs) are one of the first antibody classes mobilized during immune responses against pathogens and tumor cells. Binding to specific target antigens enables the interaction with the C1 complex which strongly activates the classical complement pathway. This biological function is the basis for the huge therapeutic potential of IgMs. But, due to their high oligomeric complexity, production, biochemical characterization, and biophysical characterization are challenging. In this study, we present recombinant production of two IgM models (IgM617 and IgM012) in pentameric and hexameric states and the evaluation of their polymer distribution using different biophysical methods (analytical ultracentrifugation, size exclusion chromatography coupled to multi-angle laser light scattering, mass photometry, and transmission electron microscopy). Each IgM construct is defined by a specific expression and purification pattern with different sample quality. Nevertheless, both purified IgMs were able to activate complement in a C1q-dependent manner. More importantly, BioLayer Interferometry (BLI) was used for characterizing the kinetics of C1q binding to recombinant IgMs. We show that recombinant IgMs possess similar C1q-binding properties as IgMs purified from human plasma.

Membrane proteins are essential bio-macromolecules involved in numerous critical biological processes and serve as therapeutic targets for a wide range of modern pharmaceuticals. Small amphipathic molecules, called detergents or surfactants, are widely used for the isolation and structural characterization of these proteins. A key requirement for such studies is their ability to maintain membrane protein stability in aqueous solution, a task where conventional detergents often fall short.

Introduction: Leukemia and radiation-induced liver toxicity are significant health challenges requiring effective therapeutic strategies. This study aimed to evaluate the therapeutic efficacy and radiosensitizing effects of Diosgenin-loaded silver nanoparticles (Dio-AgNPs) in ENU-induced leukemic mice, with a focus on their dual role in mitigating leukemia progression and γ-irradiation-induced hepatotoxicity.
Methods: Dio-AgNPs were synthesized and characterized using TEM, UV-Vis spectroscopy, FT-IR spectroscopy, and encapsulation efficiency analysis.

Aggregation of the amyloid-β peptide (Aβ) characterises and probably causes Alzheimer's disease. While lipid-mediated Aβ aggregation has been extensively studied for the 40-residue variant Aβ40, the interaction of the 42-residue variant Aβ42 with membranes has received less attention. Our time-resolved infrared spectra demonstrate that Aβ42 oligomers preserve their β-sheet structure in aqueous solution also in a membrane-mimicking environment consisting of either 1-hexadecanoyl-2-(9Z-octadecenoyl)-sn-glycero-3-phosphocholine (POPC, zwitterionic) or 1-hexadecanoyl-2-(9Z-octadecenoyl)-sn-glycero-3-phospho-(1'racglycerol) (POPG, anionic) vesicles.
