TIR-domain-containing adapter-inducing interferon-β (TRIF) is an innate immune protein that serves as an adaptor for multiple cellular signalling outcomes in the context of infection. TRIF is activated via ligation of Toll-like receptors 3 and 4. One outcome of TRIF-directed signalling is the activation of the programmed cell death pathway necroptosis, which is governed by interactions between proteins that contain a RIP Homotypic Interaction Motif (RHIM). TRIF contains a RHIM sequence and can interact with receptor interacting protein kinases 1 (RIPK1) and 3 (RIPK3) to initiate necroptosis. Here, we demonstrate that the RHIM of TRIF is amyloidogenic and supports the formation of homomeric TRIF-containing fibrils. We show that the core tetrad sequence within the RHIM governs the supramolecular organisation of TRIF amyloid assemblies, although the stable amyloid core of TRIF amyloid fibrils comprises a much larger region than the conserved RHIM only. We provide evidence that RHIMs of TRIF, RIPK1 and RIPK3 interact directly to form heteromeric structures and that these TRIF-containing hetero-assemblies display altered and emergent properties that likely underlie necroptosis signalling in response to Toll-like receptor activation.

Preterm birth and stillbirth are primary adverse pregnancy outcomes. Research during the COVID-19 pandemic revealed reductions in preterm birth in some countries, while stillbirth rates increased or remained unchanged. These findings suggest the presence of preventable risk factors associated with changes in physical activity and lower exposure to community-acquired infections due to lockdown measures, altered social interaction patterns or reduced access to antenatal care.

Varicellovirus bovinealpha (BoAHV) 1 and 5 are alphaherpesviruses that differ in their neuropathogenic potential. While BoAHV-5 causes necrotizing meningoencephalitis in calves, neurological cases associated with BoAHV-1 are less frequent. In this study we used differentiated the human neuroblastoma cell line (SH-SY5Y) to evaluate the mRNA expression of Toll-like receptor 3 (TLR3), its adaptor molecule TRIF and the production of interferons and pro-inflammatory cytokines during infection with both alphaherpesviruses.

Parasitic diseases, caused by a diverse array of parasites, remain a substantial threat to global health. Toll-like receptor 3 (TLR3) represents a pivotal element in the innate immune system, distinguished by an ability to signal via the TIR-domain-containing adapter-inducing interferon-β (TRIF)-dependent pathway upon detecting pathogen-derived double-stranded RNA (dsRNA), exosomal RNA (exoRNA), and long non-coding RNA (lncRNA). Predominantly localized on endosomal membranes, TLR3 is extensively expressed in neurons, immune cells, fibroblasts, and epithelial cells.

Purpose: To accelerate respiratory triggered free-breathing T2 mapping of the abdomen while maintaining high-quality anatomical images, accurate T2 maps, and fast reconstruction times.
Methods: We developed a flexible deep learning framework that can be trained in a fully supervised manner to improve T2-weighted images or in a self-supervised manner to reconstruct T2 maps.
Results: For retrospectively undersampled data, anatomical images and T2 maps reconstructed by the proposed deep learning method demonstrated reduced voxel-wise error compared to existing traditional and compressed sensing techniques.
