Article Abstract

CARD11 acts as a gatekeeper for adaptive immune responses after T cell or B cell antigen receptor (TCR/BCR) ligation on lymphocytes. PKCθ/β-catalyzed phosphorylation of CARD11 promotes the assembly of the CARD11-BCL10-MALT1 (CBM) complex and lymphocyte activation. Here, we demonstrated that PKCθ/β-dependent CARD11 phosphorylation also suppressed CARD11 functions in T or B cells. Through mass spectrometry-based proteomics analysis, we identified multiple constitutive and inducible CARD11 phosphorylation sites in T cells. We demonstrated that a single TCR- or BCR-inducible phosphorylation on Ser in the carboxyl terminus of CARD11 prevented the activation of the transcription factor NF-κB, the kinase JNK, and the protease MALT1. Moreover, CARD11 Ser phosphorylation sensitized BCR-addicted lymphoma cells to toxicity induced by Bruton's tyrosine kinase (BTK) inhibitors. Phosphorylation of Ser in CARD11 by PKCθ controlled the strength of CARD11 scaffolding by impairing the formation of the CBM complex. Thus, PKCθ simultaneously catalyzes both stimulatory and inhibitory CARD11 phosphorylation events, which shape the strength of CARD11 signaling in lymphocytes.

Non-remitting neutropenia in children and chronic idiopathic neutropenia (CIN) in adults have been described previously as peculiar subgroups of neutropenic patients carrying similar clinical and immunological features. The present collection comprising 25 subjects (16 adults and 9 children) mostly affected with mild (84%) and moderate (16%) neutropenia aimed to identify the underlying (possibly common) genetic background. The phenotype of these patients resemble the one described previously: no severe infections, presence of rheumathological signs, leukopenia in almost all patients and lymphocytopenia in one-third of the cohort.

Overlapping features between follicular lymphomas (FLs) and marginal zone lymphomas (MZLs) have been described, especially in their paediatric forms. Although adult primary cutaneous follicle centre lymphoma (PCFL) and primary cutaneous marginal zone lymphoma (PCMZL) of the scalp appear to display overlapping histological and molecular features, these entities have never been studied. The aim of this study was to describe the clinical, histological, and mutational profiles of PCFL and PCMZL of the scalp and to highlight possible overlapping features.

The impact of the intratumoral microbiome (ITM) on the treatment and prognosis of gastric cancer (GC) remains controversial. Our study analyzed the differential ITM in GC tissues and identified nine bacterial genera significantly associated with overall survival (OS), with seven as risk factors and two as protective factors. Three distinct clusters with varying survival outcomes were defined, demonstrating correlations with pathological stage and immune features.
