Article Abstract

The sequencing batch reactor (SBR) activated sludge process is a well-established technology for sewage treatment. One of the drawbacks of SBRs, however, total nitrogen (TN) removals is insufficient. By means of introducing four improvements, including semi-fixed biofilm carrier, sludge elevation mixing and change for the mode of influent and effluent, compliant standard for TN discharge was obtained in this novel SBR configuration during low- and high-strength sewage load. To illustrate the microbial compositions and functions of the attached biofilm on semi-fixed carrier and the suspended aggregates, as well as the nitrogen removal pathway, high throughput 16S rRNA gene amplicon sequencing, PICRUSt2 algorithm, and KEGG database were applied. The results revealed that (i) the microbial communities from suspended aggregates and biofilm samples were significantly different from each other; (ii) during low-strength sewage loads, TN removal was mainly by nitrification-denitrification. The suspended aggregates was responsible for denitrification, while the biofilm was focused on ammonium oxidation; (iii) during high-strength sewage loads, function of nitrate reductase from suspended aggregates was faded, and anammox and N assimilation by biofilm became dominant. Meanwhile, TN removal referring to the formation of L-glutamine via assimilation was the main pathway.

High processing temperatures restrict spray drying applications for heat sensitive biologics. This work highlights the potential of Phosphate Buffer Saline (PBS) as the sole stabilising excipient when spray drying large biologics via a complimentary buffer component effect. Fibrinogen (∼340 kDa) was spray dried in PBS at various temperatures and concentrations, followed by assessment of the protein's structural integrity by UV-Vis, ATR-FTIR, SEM, and PXRD analyses.

During the extraction of crude oil, water-in-oil (W/O) emulsions are mostly formed at a high pH, where water droplets can be stabilized by anionic asphaltene molecules on the surface. The study of driving forces in the electro-coalescence of these emulsions is fundamental to the efficient design of the oil dehydration process. We studied by molecular dynamics the electro-coalescence of two asphaltene-laden droplets suspended in -hexane as a model oil.

Adenosine triphosphate (ATP), a key cellular energy metabolite, has been shown to modulate protein self-assembly processes such as amyloid formation and the behavior of biological condensates through nonspecific, proteome-wide mechanisms. Gaining mechanistic insight into these effects may enable the rational use of multivalent phosphate ions as stabilizing additives for biologics. The stabilizing properties of ATP are often attributed to its hydrotropic character, arising from its combination of a nonpolar adenosine moiety and a highly charged triphosphate group.
