Article Abstract

Closed spinal dysraphism (SD) is a type of neural tube defect originating during early embryonic development whereby the neural tissue of the spinal defect remains covered by skin, often coinciding with markers of cutaneous stigmata. It is hypothesized that these events are caused by multifactorial processes, including genetic and environmental causes. We present an infant with a unique congenital midline lesion associated with a closed SD. Through comprehensive molecular profiling of the intraspinal lesion and contiguous skin lesion, an internal tandem duplication (ITD) of the kinase domain of the fibroblast growth factor receptor 1 () gene was found. This ITD variant is somatic mosaic in nature as supported by a diminished variant allele frequency in the lesional tissue and by its absence in peripheral blood. ITD results in constitutive activation of the receptor tyrosine kinase to promote cell growth, differentiation, and survival through RAS/MAPK signaling. Identification of ITD outside of central nervous system tumors is exceedingly rare, and this report broadens the phenotypic spectrum of somatic mosaic related disease.

Central nervous system tumors with BCL6 corepressor (BCOR) internal tandem duplications (ITDs) constitute a rare, recently characterized pediatric neoplasm with distinct molecular and histopathological features. To date, 69 cases have been documented in the literature, including our institutional case. These neoplasms predominantly occur in young children, with the cerebellum representing the most frequent anatomical location.

XXXXX recently added brain to its fentanyl analog testing method for 14 analogs (fluoroisobutyryl fentanyl, acetyl fentanyl, acryl fentanyl, alfentanil, butyryl fentanyl, carfentanil, fentanyl, para-fluorofentanyl, furanyl fentanyl, methoxyacetyl fentanyl, norcarfentanil, norfentanyl, sufentanil, and valeryl fentanyl) and 3 U-series drugs (U-47700, U-48800, and U-49900). Brain is a protected and isolated organ with lower metabolic activity than other tissues, which can assist in interpreting results and preserving parent drug. Limited publications testing brain samples for fentanyl and fentanyl analogs exist and none describe homogenate stability for these analytes.

Itraconazole is an oral triazole antimycotic drug. Bioequivalence studies are cornerstones for the approval of generic drug development globally. The present study describes a simple, sensitive and economical LC-MS/MS method for the determination of itraconazole and its metabolite in human plasma.

Antipyretic analgesics are typical pharmaceutical and personal care products (PPCPs) that are widely used in our daily life because they relieve fever and pain, and have anti-inflammatory and anti-rheumatic properties. These drugs inhibit the synthesis and release of prostaglandins (PGs) in the neurons of the anterior hypothalamus and exert therapeutic effects as a consequence. However, these drugs are relatively commonly misused and abused, often owing to a lack of proper medication guidance.

An ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was established to determine -(1,3-dimethylbutyl)--phenyl--phenylenediamine-quinone (6PPD-Q) in human urine and dust in order to understand the internal and external exposure levels in humans. The sample preparation conditions were systematically investigated and the chromatographic conditions and MS parameters were optimized. Briefly, internal standard C-6PPD-Q (0.
