Article Abstract

Test-samples are necessary for the development of emerging imaging approaches such as optoacoustics (OA); these can be used to benchmark new labeling agents and instrumentation, or to characterize image analysis algorithms or the inversion required to form the three-dimensional reconstructions. Alginate beads (AlBes) loaded with labeled mammalian or bacterial cells provide a method of creating defined structures of controllable size and photophysical characteristics and are well-suited for both and use. Here we describe a simple and rapid method for efficient and reproducible production of AlBes with specific characteristics and show three example applications with multispectral OA tomography imaging. We show the advantage of AlBes for studying and eventually improving photo-switching OA imaging approaches. As highly defined, homogeneous, quasi point-like signal sources, AlBes might hold similar advantages for studying other agents, light-fluence models, or the impact of detection geometries on correct image formation in the near future.

Bioprotective LAB3 cells that produce bacteriocin-like substances were entrapped in 4% (w/w) sodium alginate matrices, either with or without 10% (w/w) sodium caseinate. The effects of bead formulation-alginate alone or combined with caseinate, with or without the addition of 20% (w/w) MRS broth or M17 broth-on the culturability of LAB3 cells within the beads and their anti activity were assessed over 12 days of storage at 30 °C in closed bottles. Calcium-alginate-caseinate beads supplemented with MRS broth proved most effective in preserving both culturability and anti- activity.

Infection control and bleeding management in deep wounds remain urgent and unmet clinical challenges that demand innovative, multifunctional, and sustainable solutions. Unlike previously reported sodium alginate and silk fibroin-based gel formulations, the present work introduces a dual-functional system combining antimicrobial and haemostatic activity in the form of conformable aerogel beads. This dual-functional formulation is designed to absorb exudate, promote clotting, and provide localized antimicrobial action, all essential for accelerating wound repair in high-risk scenarios within a single biocompatible system.
