Article Abstract

Pathogens including autoantigens all failed to induce systemic lupus erythematosus (SLE). We, instead, studied the integrity of host's immune response that recognized pathogen. By stimulating TCR with an antigen repeatedly to levels that surpass host's steady-state response, self-organized criticality, SLE was induced in mice normally not prone to autoimmunity, wherein T follicular helper (Tfh) cells expressing the guanine nucleotide exchange factor DOCK8 on the cell surface were newly generated. DOCK8Tfh cells passed through TCR re-revision and induced varieties of autoantibody and lupus lesions. They existed in splenic red pulp and peripheral blood of active lupus patients, which subsequently declined after therapy. Autoantibodies and disease were healed by anti-DOCK8 antibody in the mice including SLE-model (NZBxNZW) F1 mice. Thus, DOCK8Tfh cells generated after repeated TCR stimulation by immunogenic form of pathogen, either exogenous or endogenous, in combination with HLA to levels that surpass system's self-organized criticality, cause SLE.

Unlabelled: Self-organized criticality is a hallmark of complex dynamic systems at phase transitions. Systems that operate at or near criticality have large-scale fluctuations or "avalanches", the frequency and duration power of which are best fit with a power law revealing them to be scale-free and fractal, and such power laws are ubiquitous. It is an attractive concept in neuroscience since spiking avalanches are exhibited by neural tissue, and may underpin how minuscule events could scale up to circuits and provide adaptive psychobiological function.

The immune system is an advanced, multiscale adaptive network capable of processing biological information across molecular, cellular, tissue, and systemic levels, demonstrating remarkable properties such as antifragility and criticality. We propose a unified theoretical framework based on six canonical functions-sensing, coding, decoding, response, feedback, and learning-that act as scale-invariant operational units, integrating molecular precision, collective cellular intelligence, and systemic coordination into coherent adaptive responses. Through this lens, immune function emerges from universal principles of complex network organization, including symmetry breaking, self-organized criticality, modularity, and small-world topology.

We investigate the dynamics of active paths during a two-phase flow of immiscible fluids under an external pressure drop ΔP.
