Endometrial immune response is highly associated with the homeostatic balance of the uterus and embryo development; however, the underlying molecular regulatory mechanisms are not fully elucidated. Herein, the porcine endometrium showed significant variation in mucosal immunity in proliferative and secretory phases by single-cell RNA sequencing. The loose arrangement and high motility of the uterine epithelium in the proliferative phase gave opportunities for epithelial cells and dendritic cells to cross talk with colonizing microbial community, guiding lymphocyte migration into the mucosal and glandular epithelium. The migrating lymphocytes were primarily NK and CD8 T cells, which were robustly modulated by the chemokine signaling. In the secretory phase, the significantly strengthened mechanical mucosal barrier and increased immunoglobulin A alleviated the migration of lymphocytes into the epithelium when the neuro-modulation, mineral uptake, and amino acid metabolism were strongly upregulated. The noticeably increased intraepithelial lymphocytes were positively modulated by the bacteria in the uterine cavity. Our findings illustrated that significant mucosal immunity variation in the endometrium in the proliferative and secretory phases was closely related to intraepithelial lymphocyte migration, which could be modulated by the colonizing bacteria after cross talk with epithelial cells with higher expressions of chemokine.

The senescent cell (SC) fate is linked to aging, multiple disorders and diseases, and physical dysfunction. Senolytics, agents that selectively eliminate 30-70% of SCs, act by transiently disabling the senescent cell anti-apoptotic pathways (SCAPs), which defend those SCs that are pro-apoptotic and pro-inflammatory from their own senescence-associated secretory phenotype (SASP). Consistent with this, a JAK/STAT inhibitor, Ruxolitinib, which attenuates the pro-inflammatory SASP of senescent human preadipocytes, caused them to become "senolytic-resistant".

The molecular mechanisms of these conditions are not well understood, partly due to the lack of in vitro models that mimic endometrial physiology, which limits options for targeted intervention and treatment of these diseases. Mouse models are also inadequate, as common laboratory strains do not naturally undergo a menstrual cycle comparable to that of humans.

Environmental toxicants such as maneb (MB), a dithiocarbamate pesticide, trigger progressive neuronal death, probably due to the imbalance in inflammation/resolution mechanisms, resulting in the onset of neurodegeneration. The inflammation/resolution balance is governed by G protein-coupled receptor (GPCR) signaling, but it has been poorly described in the Central Nervous System (CNS), since resolution GPCR ligands are negligible and elusive lipid compounds. These mediators are mainly synthesized by lipoxygenases (ALOX) from arachidonic acid (AA) and docosahexaenoic acid (DHA) released by specific phospholipases A2 (PLA2).

The low predictability of the effects of autologous platelet-rich plasma (PRP) in regenerative therapy for patients with type 1 and type 2 diabetes mellitus (DM) underscores the need for further research assessing the reparative effects of PRP based on the type of DM. The aim of this study was to evaluate the regenerative potential of PRP from young donors (30-40 years old) with DM1 and DM2 in vitro, specifically its effects on human dermal fibroblast cell culture. The in vitro effects of PRP from patients with type 1 and type 2 DM were investigated using a culture of human dermal fibroblasts (hTERT-HDFa) to evaluate metabolic activity, migration, proliferation of the cells, and their ability to release growth factors and exosomes.

Methods: Sweat gland secretory coils were microdissected from upper eyelid skin specimens obtained post-blepharoplasty. MECs were isolated through differential trypsinization combined with adhesion-based purification.
