Article Abstract

The human endometrium is a dynamic tissue that lines the uterus and undergoes constant remodeling, making it especially susceptible to gynecological diseases like endometriosis and endometrial cancer. The molecular mechanisms of these conditions are not well understood, partly due to the lack of in vitro models that mimic endometrial physiology, which limits options for targeted intervention and treatment of these diseases. Mouse models are also inadequate, as common laboratory strains do not naturally undergo a menstrual cycle comparable to that of humans. This study addresses this need by developing a 3D multi-compartment assembloid that mimics the architecture of endometrial tissue and recapitulates all three phases of the menstrual cycle (proliferative, secretory, and menstrual regression) within a single platform. The cellular and extracellular matrix (ECM) components in each compartment are carefully tuned based on a 3D spatial cellular map of endometrial tissue. The model contains endometrial epithelial cells enveloped in a basement membrane and endometrial stromal cells in a surrounding collagen-rich layer; this architecture allows realistic interactions between these cells and their respective ECMs. This assembloid successfully supports the controlled growth and organization of these cells, revealing reciprocal regulation of cell behavior and exhibiting compartment-specific hormonal responses, i.e., stromal decidualization. This platform enables the study of dynamic, phase-resolved, and compartment-specific paracrine signaling in human endometrial biology. By combining tissue-informed design, modular fabrication, and full-cycle hormonal responsiveness, this model sets a new benchmark for blastocyst implantation studies, organ modeling, and precision diagnostics in human reproductive health.

The Skp2-Cks1 protein-protein interaction (PPI) within the SCF ubiquitin ligase acts as a co-receptor for phosphorylated CDK inhibitors-most prominently p27-relieving CDK inhibition and advancing the cell cycle, a dependency accentuated in RB-pathway-defective cancers. Crystallographic and cryo-EM analyses delineate a composite pocket formed by the Skp2 leucine-rich-repeat groove and the phosphate-recognition site of Cks1; Cks1-centered open-closed motions further influence druggability. Using HTRF/TR-FRET and AlphaScreen biochemistry, alongside cell-based target-engagement readouts in some studies, three small-molecule classes have emerged that disrupt this PPI: 1,3-diphenyl-pyrazines and triazolo[1,5-a]pyrimidines (lead E35) with low-micromolar potency, and "Skp2E3LI" compounds with micromolar cellular activity.

Recurrent Hemothorax and Hemoperitoneum in Endometriosis: A Case Report.

Extra-pelvic endometriosis, defined as the presence of endometrial tissue or stroma outside of the pelvic cavity, is a rare cause of hemothorax and hemoperitoneum. Here, we present a case of a 34-year-old woman with a history of endometriosis who experienced recurrent, cyclical episodes of hemorrhagic pleural effusions and ascites. Despite multiple surgeries to address the ectopic endometrial implants, her symptoms persisted for years without definitive resolution, illustrating the challenges of treatment even after the diagnosis has been established.

The endometrium is a vital mucosal tissue which undergoes cyclical regeneration, differentiation, and remodeling upon hormonal, cellular, and molecular signaling networks. Dysregulation of these processes can trigger a range of pathological conditions including chronic inflammatory disorders, hyperplastic lesions, malignancies, and infertility, necessitating the need for effective therapeutic interventions. Furthermore, we are still dependent on conventional treatment modalities which are often constrained by inefficient drug biodistribution, systemic toxicity, and emergence of therapeutic resistance.
