Article Abstract

Inhibition of Mer and Axl kinases has been implicated as a potential way to improve the efficacy of current immuno-oncology therapeutics by restoring the innate immune response in the tumor microenvironment. Highly selective dual Mer/Axl kinase inhibitors are required to validate this hypothesis. Starting from hits from a DNA-encoded library screen, we optimized an imidazo[1,2-]pyridine series using structure-based compound design to improve potency and reduce lipophilicity, resulting in a highly selective probe compound . We demonstrated dose-dependent efficacy and target engagement in Mer- and Axl-dependent efficacy models using two structurally differentiated and selective dual Mer/Axl inhibitors. Additionally, efficacy was observed in a preclinical MC38 immuno-oncology model in combination with anti-PD1 antibodies and ionizing radiation.

Pancreatic cancer is a highly aggressive malignancy with a dismal prognosis, characterized by a complex tumor microenvironment that promotes immunosuppression and limits the efficacy of immune checkpoint blockade (ICB) therapy. Fibroblast activation protein (FAP) is overexpressed in the tumor stroma and represents a promising target for therapeutic intervention. Here, we developed a novel antibody-drug conjugate (ADC) targeting FAP, and investigated its anti-tumor activity and ability to enhance ICB efficacy in pancreatic cancer.

Giant two-photon upconversion from 2D exciton in doubly-resonant plasmonic nanocavity.

Photon upconversion through high harmonic generation, multiphoton absorption, Auger recombination and phonon scattering performs a vital role in energy conversion and renormalization. Considering the reduced dielectric screening and enhanced Coulomb interactions, semiconductor monolayers provide a promising platform to explore photon upconversion at room temperature. Additionally, two-photon upconversion was recently demonstrated as an emerging technique to probe the excitonic dark states due to the extraordinary selection rule compared with conventional excitation.

Purpose: This study evaluated how the formation of various types of precipitates affects the accuracy of electronic apex locators in determining the working length.

Methods: Two hundred and forty extracted human maxillary incisors were selected. A total of eight groups were used: four groups (n = 30) for the application of different irrigants; saline, sodium hypochlorite (5.

Objectives: Despite efforts to promote mental health in the workplace, employee mental health challenges continue to increase. Organizations continue to face significant challenges to support employees in safely returning to work after mental health-related leaves and to ensure long-term stability without relapse. This literature-based study was performed to clarify support skills that occupational health nurses employ to facilitate return to work for employees with mental health issues.

The microbiome and volatile organic compounds reflecting the state of decomposition in an indoor environment.

Given that a variety of factors can affect the decomposition process, it can be difficult to determine the post-mortem interval (PMI). The process is highly dependent on microbial activity, and volatile organic compounds (VOCs) are a by-product of this activity. Given both have been proposed to assist in PMI determination, a deeper understanding of this relationship is needed.
