The ligand of numb-protein X1 () acts as a proto-oncogene by inhibiting stability; however, the regulation of expression has not been investigated. In this study, we screened chemicals to identify factors that potentially regulate expression. We found that expression levels were decreased by DNA damage, including that by cisplatin. Upon treatment with lipopolysaccharide (LPS) and phorbol 12-myristate 13-acetate (PMA), expression levels increased. In addition, cell-cycle progression increased upon expression; the levels of S and G2/M populations were correlated with expression. Moreover, in CRISPR-Cas9-mediated knockout cells, we observed a delay in cell-cycle progression and a downregulation of genes encoding the cell-cycle markers cyclin D1 and cyclin E1. Finally, the upregulation of -activated cell-cycle progression and increased resistance to cisplatin-mediated cell death. Taken together, these results suggest that contributes to cell-cycle progression and cisplatin resistance.

Primary microcephaly, a rare congenital condition characterized by reduced brain size, occurs due to impaired neurogenesis during brain development. Through whole-exome sequencing, we identified compound heterozygous loss-of-function mutations in CENTRIN 3 (CETN3) in a 5-year-old patient with primary microcephaly. As CETN3 has not been previously linked to microcephaly, we investigated its potential function in neurodevelopment in human pluripotent stem cell-derived cerebral organoids.

Translocon-associated protein subunit beta (TRAPβ), also known as signal sequence receptor 2 (SSR2) serves as an auxiliary protein facilitating co-translational translocation in the endoplasmic reticulum (ER); however, its role in colorectal cancer is unknown to date. The objectives of the current study are to examine if TRAPβ/SSR2 knockdown affects the cell proliferation and to elucidate mechanisms by which TRAPβ/SSR2 regulates proliferation of human colorectal cancer.
