Article Abstract

Golgi reassembly stacking proteins (GRASPs) play important roles in Golgi structure formation, ER stress response, and unconventional secretion in eukaryotic cells. However, GRASP functions in oomycetes haven't been adequately characterized. Here, we report the identification and functional analysis of , a GRASP-encoding gene from the soybean-infecting oomycete . Transcriptional profiling showed that expression is up-regulated at the infection stages. knockout mutants were created using the CRISPR/Cas9 system. These mutants exhibited impaired vegetative growth, zoospore release and virulence. PsGRASP was involved ER stress responses and altered laccase activity. Our work suggests that PsGRASP is crucial for development and pathogenicity.

The Golgi apparatus undergoes systematic disassembly and reassembly during the cell cycle, a process requiring membrane fusion mediated by the AAA+ ATPase p97/VCP and its adaptor p47. While the p97-p47 complex plays a pivotal role in post-mitotic Golgi reassembly, the exact molecular mechanism underlying its function has not been completely understood. In particular, the conformational flexibility and dynamic feature of p47 hinders its structural characterization by cryo-electron microscopy and X-ray crystallography.

The Golgi complex serves as a major noncentrosomal microtubule-organizing center in animal cells, with Golgi-derived microtubules playing a crucial role in Golgi assembly, structural organization, cell migration, and various Golgi-related cellular functions. In this study, we identify GAS2L1 as a key regulator of Golgi-based microtubule growth and Golgi reassembly. Loss of GAS2L1 leads to significant defects in Golgi morphology and impairs nocodazole washout-induced Golgi reassembly.

Primordial follicle pool is the foundation of female reproductive life and abnormal primordial follicle activation may lead to severe diseases such as premature ovarian failure and premature ovarian insufficiency. Golgi reassembly stacking protein 2 (GORASP2) plays an important role in autophagy by regulating autophagy maturation through glycosylation modification. In the current study we found that GORASP2 is a key factor in mammalian primordial follicle activation through autophagy lysosome pathway.

Improving treatment options for head and neck squamous cell carcinoma (HNSCC) requires a deeper understanding of the tumor microenvironment, particularly cancer-associated fibroblasts (CAFs). We previously reported that HNSCC-derived FGF2/bFGF (fibroblast growth factor 2) triggers cytokine release from CAFs via secretory autophagy. Here, using transmission electron microscopy, live-cell imaging, and immunofluorescence, we show that CAF autophagosomes transport cargo, including IL6, to the plasma membrane for secretion.
