The cellular response to interferon (IFN) is essential for antiviral immunity, IFN-based therapy and IFN-related disease. The plasma membrane (PM) provides a critical interface between the cell and its environment, and is the initial portal of entry for viruses. Nonetheless, the effect of IFN on PM proteins is surprisingly poorly understood, and has not been systematically investigated in primary immune cells. Here, we use multiplexed proteomics to quantify IFNα2a-stimulated PM protein changes in primary human CD14+ monocytes and CD4+ T cells from five donors, quantifying 606 and 482 PM proteins respectively. Comparison of cell surface proteomes revealed a remarkable invariance between donors in the overall composition of the cell surface from each cell type, but a marked donor-to-donor variability in the effects of IFNα2a. Furthermore, whereas only 2.7% of quantified proteins were consistently upregulated by IFNα2a at the surface of CD4+ T cells, 6.8% of proteins were consistently upregulated in primary monocytes, suggesting that the magnitude of the IFNα2a response varies according to cell type. Among these differentially regulated proteins, we found the viral target Endothelin-converting enzyme 1 (ECE1) to be an IFNα2a-stimulated protein exclusively upregulated at the surface of CD4+ T cells. We therefore provide a comprehensive map of the cell surface of IFNα2a-stimulated primary human immune cells, including previously uncharacterized interferon stimulated genes (ISGs) and candidate antiviral factors.

Merocyanine photoacids (MCHs) have found applications in chemical, material, energy, and biomedical areas, and are currently being investigated for industrial applications. Hydrolysis, relatively high dark acidity, and moderate solubility in water are the major concerns for their practical applications. View Article and Find Full Text PDFThis review analyzes Russian and international literature on the treatment of bilateral limbal stem cell deficiency (LSCD), focusing on the use of Simple Oral Mucosal Epithelial Transplantation (SOMET) as a surgical method for restoring the ocular surface. Contemporary sources report 64 cases of SOMET used in the treatment of bilateral LSCD: 35 cases of chemical burns, 16 of thermal burns, 7 cases of Stevens-Johnson syndrome, 1 keratitis, 1 cicatricial pemphigoid, 1 dermoid, 1 case of drug-induced LSCD (mitomycin C), etc. Notably, all transplantations resulted in complete epithelialization, and in 3 cases, penetrating keratoplasty was subsequently performed with favorable functional and anatomical outcomes.

Two major protein recycling pathways have emerged as key regulators of enduring forms of synaptic plasticity, such as long-term potentiation (LTP), yet how these pathways are recruited during plasticity is unknown. Phosphatidylinositol-3-phosphate (PI(3)P) is a key regulator of endosomal trafficking and alterations in this lipid have been linked to neurodegeneration. Here, using primary hippocampal neurons, we demonstrate dynamic PI(3)P synthesis during chemical induction of LTP (cLTP), which drives coordinate recruitment of the SNX17-Retriever and SNX27-Retromer pathways to endosomes and synaptic sites.

The mechanical properties of graphene are investigated using classical molecular dynamics simulations as a function of temperature T and external stress τ. The elastic response is characterized by calculating elastic constants via three complementary methods: (i) numerical derivatives of stress-strain curves, (ii) analysis of cell fluctuation correlations, and (iii) phonon dispersion analysis. Simulations were performed with two interatomic models: an empirical potential and a tight-binding electronic Hamiltonian.
