The incidence of myelodysplastic syndrome increases with aging and the early diagnosis enables optimal care of these diseases. The DxH 800 hematology analyzer measures and calculates 126 cytological parameters, but only 23 are used for routine CBC assessment. The goal of this study was to use the 103 unexploited "research parameters" to develop an algorithm allowing for an early detection of subclinical MDS patients by triggering morphological analysis. Blood sample parameters from 101 MDS patients and 88 healthy volunteers were analyzed to identify the critical "research parameters" with: (i) the most significant differences between MDS patients and healthy volunteers, (ii) the best contributions to principal component analysis (PCA), first axis, and (iii) the best correlations with PCA, first two axes (cos > 0.6). Ten critical "research parameters" of white blood cells were identified, allowing for the calculation of an MDS-likelihood score (MDS-LS), based on logistic regression. Automatic calculation of the MDS-LS is easily implementable on the middleware system of the DxH 800 to generate a flag for blood smear review, and possibly early detection of MDS patients in the general population.

Background: GBA1 variants are the major genetic risk factor for Parkinson's Disease (PD) and account for 5-30% of PD cases depending on the population and age at onset of the disease.
Objectives: The aim of this study was to assess whether Artificial Intelligence (AI) could predict GBA1-mutated genotype in PD (GBA1-PD). Particularly, the main objective was to identify a Machine Learning (ML) model capable of accurately providing a pre-test estimate of GBA1-mutated status, relying on the clinical and demographic variables with the highest predictive value.

Background: Standard treatments for advanced cervical cancer, such as paclitaxel-cisplatin combination (TP) chemotherapy, are often limited by reduced efficacy and significant toxicity. Cinobufacini (Huachansu), a traditional Chinese medicine, has demonstrated potential in enhancing the effectiveness of conventional cancer therapies.
Methods: A systematic search of Web of Science, PubMed, Cochrane, Embase, China National Knowledge Infrastructure, and other databases was conducted up to July 30, 2024.
Paroxysmal Nocturnal Hemoglobinuria (PNH) clones are frequently found in hypoplastic myelodysplastic syndromes (hMDS), though less commonly than in aplastic anemia. In contrast, the coexistence of hemolytic PNH with large clones and classical, hypercellular MDS (non-hMDS) is rare and likely underrecognized in clinical practice. Since 2014, 229 MDS patients have been seen at our department.

Chronic myelomonocytic leukemia (CMML) is an aggressive hematologic neoplasm characterized by an expansion of CD123 monocytes and plasmacytoid dendritic cells (pDCs). pDC bone marrow clusters in CMML have been associated with higher rates of acute myeloid leukemia transformation. We evaluated tagraxofusp, a CD123-targeted therapy, in a phase 1/2 trial for patients with CMML.
