Article Abstract

The human intestinal stem cell niche supports self-renewal and epithelial function, but little is known about its development. We used single-cell mRNA sequencing with in situ validation approaches to interrogate human intestinal development from 7-21 weeks post conception, assigning molecular identities and spatial locations to cells and factors that comprise the niche. Smooth muscle cells of the muscularis mucosa, in close proximity to proliferative crypts, are a source of WNT and RSPONDIN ligands, whereas EGF is expressed far from crypts in the villus epithelium. Instead, an PDGFRA/F3/DLL1 mesenchymal population lines the crypt-villus axis and is the source of the epidermal growth factor (EGF) family member NEUREGULIN1 (NRG1). In developing intestine enteroid cultures, NRG1, but not EGF, permitted increased cellular diversity via differentiation of secretory lineages. This work highlights the complexities of intestinal EGF/ERBB signaling and delineates key niche cells and signals of the developing intestine.

Inherited genetic variants contribute to Barrett's esophagus (BE) and esophageal adenocarcinoma (EAC), but it is unknown which cell types are involved in this process. We performed single-cell RNA sequencing of BE, EAC, and paired normal tissues and integrated genome-wide association data to determine cell-type-specific genetic risk and cellular processes that contribute to BE and EAC. The analysis reveals that EAC development is driven to a greater extent by local cellular processes than BE development and suggests that one cell type of BE origin (intestinal metaplasia cells) and cellular processes that control the differentiation of columnar cells are of particular relevance for EAC development.

Recent years have seen advances in clinical biochemistry of domestic animals which have highlighted comparative differences between species and have also identified fundamental aspects of the biochemical mechanisms in physiological conditions and disease, that have implications across species, including human, health and welfare. From investigations in diverse species using biochemical, immunological, proteomic and metabolomic approaches a series of species particularities and unexpected results for some biomarkers have been made. These observations cover (1) the differences between species in the acute phase protein (APP) response to infection and inflammation; (2) the non-hepatic synthesis and release in the mammary gland, adipose tissue and intestine of APP (3) the response of haptoglobin (HP) as a biomarker for stress; (4) observations in non-mammalian species related to hemopexin and HP; (5) the response of bile acids in milk to mastitis; (6) barley serine protease inhibitors being identified in bovine faeces; (7) alkaline phosphatase being present in bovine nasal secretion; (8) saliva findings with analytes such as adenine deaminase showing different activity between saliva and serum and a detergent-like surfactant protein, latherin being found in equine saliva and sweat and (9) serum enzymes and selective muscle protein reaction of Atlantic salmon as an example of the differences in biochemistry between terrestrial and aquatic species.
