The anticoagulant proteins antithrombin (AT), protein C (PC), protein S (PS), and tissue factor pathway inhibitor (TFPI) are physiological regulators of the blood coagulation pathway and have complex roles in causing or modifying the severity of bleeding and clotting disorders. The anticoagulant activity, as well as alternative physiological functions, of these proteins also impact many other diseases beyond bleeding and clotting disorders. We searched PubMed for articles published between 2018 and early 2025 that pertained to alternative functions of these physiological anticoagulants in clinical disease and selected therapeutic applications in the following categories: cancer, cardiovascular disease, novel therapeutics, pregnancy health, inflammation and infection, blood-brain barrier protection, stroke, novel developments in thrombosis, and endothelial and platelet interactions.

Introduction: The infection caused by the COVID-19 virus is associated with thromboembolic events and severe inflammatory reactions, significantly impacting the prognosis of infected patients. Numerous studies have indicated that COVID-19 patients often exhibit a hypercoagulable state, disseminated intravascular coagulation, and overwhelming inflammation, particularly in critically ill patients with multiple comorbidities requiring admission to the ICU. This study aims to assess the prognostic significance of alterations in coagulation, inflammatory, and blood chemistry markers in COVID-19 patients both before and during admission to the ICU.

Partial atrioventricular septal defect (AVSD) is a congenital heart anomaly that typically manifests in childhood and rarely presents in elderly patients. However, this anomaly can lead to hemodynamically significant cardiopulmonary complications when left untreated. In this report, we present a case of a 68-year-old male patient who presented with symptoms of heart failure and atrial flutter and was found to have partial AVSD incidentally discovered by echocardiography.

Background: Post-infectious glomerulonephritis (PIGN) is one of the leading causes of acute nephritis in children worldwide. C3 glomerulopathy (C3G) is a rare form of membranoproliferative glomerulonephritis (MPGN) characterised by either genetic or acquired dysregulation of the alternative complement pathway resulting in predominant C3 deposition within the glomeruli. The overlap between atypical post-streptococcal glomerulonephritis (PSGN), a subset of PIGN primarily induced by streptococcal species, and C3G has attracted considerable attention in recent clinical trials.
