Protein-protein interactions (PPIs) contribute to the onset and/or progression of several diseases, especially cancer, and this discovery has paved the way for considering disruption of the PPIs as an attractive anti-tumor strategy. In this regard, simple and efficient biophysical methods for detecting the interaction of the inhibitors with the protein counterpart are still in high demand. Herein, we describe a convenient NMR method for the screening of putative PPI inhibitors based on the use of "hot peptides" (HOPPI-NMR). As a case study, HOPPI-NMR was successful applied to the well-known p53/MDM2 system. Our outcomes highlight the main advantages of the method, including the use of a small amount of unlabeled proteins, the minimization of the risk of protein aggregation, and the ability to identify weak binders. The last leaves open the possibility for application of HOPPI-NMR in tandem with fragment-based drug discovery as a valid strategy for the identification of novel chemotypes acting as PPI inhibitors.

This study investigates the effects of L-carnitine on nuclear maturation and fertilization in cattle and goat oocytes. Ovaries were collected from females with poor reproductive efficiency in the tropical climate, and cumulus-oocyte complexes (COCs) were retrieved from large antral follicles. COCs were cultured with varying concentrations of L-carnitine (0, 0.

Plasmablastic lymphoma (PBL) is a rare and aggressive non-Hodgkin lymphoma with a poor prognosis and short survival rates. It is classified as a large B-cell lymphoma subtype, but carries a plasmacytic immunophenotype. Therefore, PBL has pathogenetic overlaps with diffuse large B-cell lymphoma not otherwise specified (DLBCL NOS) and plasma cell neoplasms (PCNs).

Background: The proteome is a valuable resource for pinpointing therapeutic targets. Therefore, we conducted a proteome-wide Mendelian randomization (MR) study aimed at identifying potential protein markers and therapeutic targets for Anti-N-Methyl-D-Aspartate Receptor Encephalitis (NMDAR-E).
Methods: Protein quantitative trait loci (pQTLs) were obtained from seven published genome-wide association studies (GWASs) focusing on the plasma proteome, resulting in summary-level data for 734 circulating protein markers.

Background: While associations between psychiatric disorders and hypothyroidism have been observed, the underlying causal relationships remain unclear. Exploring the causal relationships among them can provide new insights into the underlying mechanisms.
Methods: Two-sample Mendelian randomization (MR) was conducted to assess the causal effects of seven psychiatric disorders on hypothyroidism.

Programmed cell death (PCD), which describes cell death regulated by a sequence of gene expression events, strongly impacts the prognosis of spinal cord injury (SCI). Nevertheless, the connections between the various PCD types and the cross-linked genes regulate that these types of cell death in SCI remain unclear. This study sought to identify and investigate the key genes connections that regulated PCD in SCI.
