Cells subjected to stress situations mobilize specific membranes and proteins to initiate autophagy. Phosphatidylinositol-3-phosphate (PI3P), a crucial lipid in membrane dynamics, is known to be essential in this context. In addition to nutriments deprivation, autophagy is also triggered by fluid-flow induced shear stress in epithelial cells, and this specific autophagic response depends on primary cilium (PC) signaling and leads to cell size regulation. Here we report that PI3KC2α, required for ciliogenesis and PC functions, promotes the synthesis of a local pool of PI3P upon shear stress. We show that PI3KC2α depletion in cells subjected to shear stress abolishes ciliogenesis as well as the autophagy and related cell size regulation. We finally show that PI3KC2α and VPS34, the two main enzymes responsible for PI3P synthesis, have different roles during autophagy, depending on the type of cellular stress: while VPS34 is clearly required for starvation-induced autophagy, PI3KC2α participates only in shear stress-dependent autophagy.

Background: The objective of this study was to compare the effects of daily consumption of white potatoes compared with white rice on cardiometabolic health in individuals with type-2 diabetes (T2D).
Objective: To determine the effects of white potato consumption compared to white rice (a commonly consumed refined grain) on indices of glycemic control and cardiovascular health in individuals with overweight or obesity and T2D.
Methods: In this randomized crossover study, comparative control trial, 24 adults with T2D [45-80 y, body mass index (kg/m) 25-40] consumed baked white potatoes (100 g) or calorie-matched white rice (75 g) daily for 12 wk, separated by a 2-wk washout, with assessments of glycemic control, lipids, inflammation, blood pressure, endothelial function, and body composition at baseline (only 1 baseline visit included as a covariate in statistical analyses), 6 wk, and 12 wk.

The screw-retained implant-supported crown is a durable, aesthetic restoration, but debonding between the crown and abutment remains a challenge to survivability. The purpose of this work was to devise an abutment shape that can be embedded into the crown while the crown is being additively manufactured. The result was a mechanically retained, no-adhesive abutment and crown unit that is mounted to the implant fixture.

Triply periodic minimal surfaces have garnered significant interest in the field of biomaterial scaffolds due to their unique structural properties, including a high surface-to-volume (S/V) ratio, tunable permeability, and the potential for enhanced biocompatibility. Bone scaffolds necessitate specific features to effectively support tissue regeneration. This study examines the permeability and active cell proliferation area of advanced Triply Periodic Minimal Surface (TPMS) lattice structures, focusing on a novel lattice design.

The usability and versatility of autoinjectors in managing chronic and autoimmune diseases have made them increasingly attractive in medicine. However, investigations into autoinjector designs require an understanding of the kinematic properties and fluid behaviour during injection. To optimise injection efficiency, this study develops a mathematical and computational fluid dynamics (CFD) model of an IM autoinjector by investigating the effects of viscosity, needle length, needle diameter, and medication volume on the injection process.

Sectionally nonlinearly functionally graded (SNFG) structures with triply periodic minimal surface (TPMS) are considered ideal for bone implants because they closely replicate the hierarchical, anisotropic, and porous architecture of natural bone. The smooth gradient in material distribution allows for optimal load transfer, reduced stress shielding, and enhanced bone ingrowth, while TPMS provides high mechanical strength-to-weight ratio and interconnected porosity for vascularization and tissue integration. Wherein, The SNFG structure contains sections with thickness that varies nonlinearly along their length in different patterns.
