Article Abstract

Presequence protease (PreP) is a proteostatic enzyme that plays a key role in the maintenance of mitochondrial health. Defects in PreP stability are associated with neurological disorders in humans, and altered activity of this enzyme modulates the progress of Alzheimer's disease-like pathology in mice. As agonists that boost PreP proteolytic activity represent a promising therapeutic avenue, we sought to determine the structural basis for the action of benzimidazole derivatives (3c and 4c), first reported by Vangavaragu et al. (Eur. J. Med. Chem. 76 (2014) 506-516) that enhance the activity of PreP. However, we found the published procedure for the synthesis of 3c yielded aldimine A instead. We then developed an alternative synthesis and obtained 3c, termed compound C, and an alternative benzimidazole derivative, termed compound B. We tested compounds A, B and C for their ability to enhance the activities of human PreP. In contrast to the previous report, we observed that none of the compounds A, B, or C (3c) modulated the catalytic activity of human PreP. Here we report our findings on the mis-identification of the reported benzimidazoles and the lack of biological activity of such compounds on human PreP. Thus, PreP modulators for PreP-based therapies remain to be discovered.

Despite growing recognition of the interdependencies of resilience across systems, sectors, and levels (SSLs), translating this understanding into coordinated action remains a challenge. This study identifies seven systemic gaps that reinforce a persistent know–do gap, creating an unhealthy that reinforces fragmentation across SSLs. In response, seven prerequisites for synergizing resilience are proposed, along with a working definition of Synergistic Resilience.

Introduction: Oral HIV pre-exposure prophylaxis (PrEP) is a highly effective biomedical intervention for HIV prevention, but its access and utilisation are challenging, especially in high-burden settings such as Kenya. For potential PrEP users, long delays and repeated consultations with several providers are obstacles to both PrEP uptake and continuation. Nondisclosure of use of antiretroviral medications in Canadian blood donors.

Introduction: Donors are deferred if they are on antiretroviral medications (ARV) as post-exposure or pre-exposure prophylaxis (PEP or PrEP) for human immunodeficiency virus (HIV). We assessed donor compliance by measuring ARV levels in selected anonymized donor samples collected from September 22, 2022 to December 31, 2024, almost all after the introduction of sexual risk behavior screening.

Methods: EDTA plasma samples collected at the time of donation (retention samples) were retrieved, frozen, and shipped for measurement of tenofovir and emtricitabine.

Prolyl endopeptidase (PREP) drives neurodegenerative diseases through dual mechanisms involving enzymatic activity and protein-protein interactions (PPIs), yet current inhibitors predominantly target single pathways. Prolyl endopeptidase (PREP) fuels neurodegeneration via enzymatic cleavage and pathological PPIs, yet current inhibitors usually target only one facet. In this study, leveraging our developed high-sensitivity and high-specificity near-infrared fluorescent probe Z-GP-ACM, we established and validated a screening platform for PREP inhibitors with mouse brain S9 instead of the human recombinant PREP.
