Article Abstract

Spectrophotometric analysis of metabolic enzyme activity from homogenized tissues is a valuable method for investigating mitochondrial content and capacity. Enzyme activity is normally measured in single cuvette spectrophotometers, requiring a large sample volume and low throughput. Here, we describe microplate assays for high-throughput analysis of mitochondrial enzymes citrate synthase, β-hydroxyacyl CoA dehydrogenase, aconitase, and mitochondrial electron transport system (ETS) complexes I, II, III, and IV.

Mycobacteriophage-encoded LysinB enzymes target mycolyl ester linkages in mycolyl-arabinogalactan-peptidoglycan of mycobacterium hosts and generally exhibit a globular architecture. Here, we present the structural and functional characterization of a novel Mycobacterium fortuitum prophage-encoded modular LysinB (LysinB_MF), which contains the α/β hydrolase domain and a distinct peptidoglycan-binding domain (PGBD). The enzyme's active site features the conserved Ser-Asp-His catalytic triad common to esterases and forms a funnel-like topology.

Histomonas meleagridis is a parasitic protozoan which causes histomoniasis (blackhead disease) in a wide range of birds, including domesticated chickens and turkeys, representing a significant health problem in avian veterinary medicine. Despite being classified as an anaerobic parasite, H. meleagridis can survive transient exposure to oxygen while little is known about the mechanisms that allow this organism to cope with exposure to varying oxygen levels.
