To develop a detection method for single nucleotide polymorphisms (SNPs) of bronchial asthma (BA) susceptibility genes (, and ) based on fluorescence PCR melting curves. Peripheral blood samples from 33 patients with BA were collected. DNA was extracted, and positive plasmids were constructed. Probes and primers for fluorescence polymerase chain reaction (PCR) were designed according to and sequences, and the SNPs were separately detected by gene sequencing and fluorescence PCR melting curve. The system was successfully divided into 3 SNPs, including , and , and a comparison of sequencing methods showed that the results were completely consistent. The lowest detection limit was 1 ng/reaction, the sensitivity and specificity were 100%, and this method had high repeatability (CV = 2.8%). The fluorescence PCR melting curve method is suitable for the rapid and accurate classification of SNPs. The method is economical, simple, and efficient, and is suitable for the screening of the susceptible gene SNPs in a large-scale population of patients with BA.

MicroRNAs (miRNAs) are considered more stable than mRNA, but the impact of progressive thawing of biological samples after freezing as may happen during shipping delays has not been quantified. To address this, we utilized digital PCR to estimate the absolute concentrations of select miRNAs following progressive thawing of human plasma and maintenance at ambient temperature. Specifically, we quantified let-7b-3p, miR-144-5p, miR-150-5p, miR-517a-3p, miR-524-5p, and miR-1283, which have varying abundance in plasma.

Objectives: To investigate the impact of prenatal fear stress on placental amino acid transport and emotion and cognition development in offspring rats.
Methods: Thirty pregnant Wistar rats were randomized equally into control and fear stress (induced using an observational foot shock model) groups. In each group, placental and serum samples were collected from 6 dams on gestational day 20, and the remaining rats delivered naturally and the offspring rats were raised under the same conditions until 8 weeks of age.

The global emergence of SARS-CoV-2 has highlighted the need for rapid, sensitive, and affordable diagnostic tools, not only for human health but also for animal surveillance within a One Health framework. This study aimed to evaluate the performance of a SYBR Green-based real-time quantitative PCR (qPCR) assay for the detection of SARS-CoV-2 from animal samples, focusing on domestic dogs and cats. A total of 140 oropharyngeal swab samples were collected and analyzed using primers targeting a 139-bp fragment of the N gene of SARS-CoV-2.
