Engrailed-2 (EN2) is a homeodomain-containing transcription factor that has roles in boundary formation and neural guidance in early development, but which is also expressed in a range of cancers. In addition to transcriptional regulation, it is secreted by cells and taken up by others through a mechanism that is yet to be fully elucidated. In this study, the distribution of EN2 protein in cells was evaluated using immunofluorescence with a set of antibodies raised against overlapping epitopes across the protein, and through the use of an EN2-GFP construct. MX2 expression in primary prostate tumors was evaluated using immunohistochemistry. We showed that EN2 protein is present in the cell membrane and within microvesicles that can be secreted from the cell and taken up by others. When taken up by normal cells from the stroma EN2 induces the expression of MX2 (MxB), a protein that has a key role in the innate immune response to viruses. Our findings indicate that EN2 secretion by tumors may be a means of preventing viral-mediated immune invasion of tissue immediately adjacent to the tumor.

Glioblastoma (GBM) is the most aggressive primary brain tumour, with limited treatment options and a propensity for rapid development of resistance to therapies. Previous studies have indicated that fatty acid metabolic reprogramming is a critical marker of tumour progression and plays a significant role in the proliferation and migration of cancer cells. However, research on fatty acid synthesis metabolism in GBM is relatively limited, and the underlying mechanisms warrant further investigation.

Objective: To investigate the impact of drinking different caloric sweeteners from immaturity to young adulthood on hypothalamic controlled physiological functions and hypothalamic global gene expression using a rat model.
Methods: Young female Sprague-Dawley rats (age 28 days) were randomly assigned ( = 7 rats/group) to drink water sweetened with 13% (w/w) sugar as either high fructose corn syrup (HFCS), sucrose, fructose, or water (control) for 8 weeks. Hypothalamic controlled physiological function measurements included: energy intake, stress, and estrous cycles.

Engrailed-2 (EN2) protein, a transcription factor in the homologous domain expressed in prostate cancer (PCa) cells and secreted into the urine, is considered a promising biomarker for noninvasive detection of PCa. EN2 protein in urine samples can be obtained by noninvasive means, but the low biomarker concentration in urine samples poses a great challenge for noninvasive detection of the PCa biomarker. Herein, we develop a solution-gated graphene transistor (SGGT) biosensor to detect the biomarker of the EN2 protein for PCa diagnosis.

Objective: This study aims to demonstrate that the EN2 protein in cervical mucus may serve as a novel biomarker for screening endometrial cancer.
Materials And Methods: This study included 133 patients who were treated at Beijing Obstetrics and Gynecology Hospital. According to the pathological results of hysteroscopy endometrial biopsy, the patients were divided into endometrial cancer group (n = 55), endometrial atypical hyperplasia group (n = 16), benign lesion group (n = 28), and control group (n = 34).
