Eleutherococcus extract mixture (EEM) is an herbal mixture of dried stem of and germinated barley, which has been highly effective, in previous screening and among the traditional medicines to tonify innate and acquired , respectively. In this study, we investigate the effects of EEM on endochondral bone formation. Female adolescent rats were given EEM, growth hormone or vehicle for 10 days. Tetracycline was intraperitoneally injected to light the fluorescent band 72 h before sacrifice to determine endochondral bone formation. In order to evaluate endocrine or paracrine/autocrine mechanisms, expressions of insulin-like growth factor 1 (IGF1), insulin-like growth factor binding protein 3 (IGFBP3), or bone morphogenetic protein 2 (BMP2) were evaluated after EEM administration in liver or growth plate (GP). EEM oral administration significantly increased endochondral bone formation and proliferative and hypertrophic zonal heights of tibial GP. EEM also upregulated hepatic IGF1 and IGFBP3 mRNA expressions, and IGF1 and BMP2 expressions in GP. Taken together, EEM increases endochondral bone formation through stimulating proliferation and hypertrophy with upregulation of hepatic IGF1 and IGFBP3 expressions. Considering immunohistochemical studies, the effect of EEM may be due to increased local IGF1 and BMP2 expression in GP, which may be considered growth hormone (GH)-dependent endocrine and autocrine/paracrine pathways.

The mechanisms mediating endochondral bone formation remain incompletely understood. Here, we show that CXXC Finger Protein 1 (CFP1) is required for the onset of chondrogenesis during forelimb development. CFP1-deficient mesenchymal progenitor cells (LMPs) retain an immature molecular signature with elevated FGF and SHH signaling and repressed BMP signaling, in part, due to (1) reduced expression of type I BMP receptors, (2) reduced Smad1 protein levels and (3) an altered extracellular niche.

In fish, epibranchials are dorsal facial skeletal elements composing the branchial basket, which articulate with ceratobranchials ventrally and pharyngobranchials dorsally. They form in the posterior pharyngeal arches through endochondral bone formation. In zebrafish, the development of epibranchial structures has not been described in detail at the genetic and cellular levels compared to the development of the jaw skeleton.

The human sphenoid bone (SB), centrally located at the cranial base, is structurally and developmentally complex. This study presents a comprehensive framework combining Selective Laser Melting (SLM) of Titanium (Ti64) alloys, finite element simulation, and artificial intelligence (AI) to advance orthopedic implants' design and predictive evaluation. Dense Ti64 specimens are fabricated using ten distinct SLM parameter sets to explore the effects of volumetric energy density (VED) on mechanical behavior, porosity distribution, and microstructural integrity. Optimal VED ranges are identified to balance defect minimization and mechanical performance, with porosity levels strongly influencing tensile strength and Young's modulus.

Background: Identification of biomarkers to predict the risk of healing delays are of huge clinical interest since 10% of fracture patients progress to delayed or non-union. During endochondral ossification, which takes place in mechanically unstable regions, the bone regenerates through a cartilage intermediate. We previously identified miR-1246, miR-335-5p and miR-193a-5p as fracture-related biomarkers in patient serum, but they appear not to have a functional role in an in vitro model of direct ossification.
