A simple and fast immunoassay strip to detect Acidovorax citrulli (Ac) using fluorescein isothiocyanate as a marker was developed. Fluorescein isothiocyanate (FITC) was added to sample culture medium for bacteria incubation, and the bacteria could emit a yellow-green fluorescence under ultraviolet light and become a fluorescent probe. This immunofluorescence strip (IFS) was based on the binding between fluorescent bacteria and the unlabeled monoclonal antibody (McAb) immobilized on the test area in nitrocellulose membrane. The detection limit of the strip was 10 CFU/ml with a result that could be observed within 10 min. The IFS could detect eight strains of Ac and display no cross-reactions with 30 other pathogenic strains. The detection results would not be affected by impurities in plant or unknown microorganisms in natural field samples and were consistent with PCR results, indicating that the IFS has high accuracy. This is the first report of using only one unlabeled McAb to develop a direct-type immunofluorescence strip for the rapid detection of Ac. The IFS reduced detection time and simplified operation procedures compared with the traditional enzyme-linked immunosorbent assay (ELISA) and PCR methods. In addition, this simple and inexpensive method will play a significant role in monitoring plant pathogens on field detection.

Background: Lysosomes serve not only in the degradation of cellular components but also as calcium (Ca) stores. In this study, we investigated the effects of trans-Ned19, an inhibitor of lysosomal calcium channels known to block two-pore channels (TPCs), on fertilization and oocyte activation in mice.
Methods: Pronuclear formation was assessed via Hoechst 33342 staining, cortical granule release was evaluated using agglutinin-fluorescein isothiocyanate (LCA-FITC) staining, intracellular Ca levels were monitored with Cal-520 AM, and sperm motility was analyzed using a sperm motility analysis system (SMAS).
Specific protein detection plays a crucial role in biological analysis and clinical diagnostics, serving as an essential tool for disease diagnosis, therapeutic monitoring, and biological research. However, conventional methods such as immunofixation electrophoresis (IFE) and western blotting (WB) suffer from complex workflows, time-consuming operations, and limited quantification capabilities owing to intricate staining and de-staining procedures. In addition, these traditional immunological detection methods require extensive manual handling and specialized expertise, while low levels of automation restrict their applicability to high-throughput or large-scale analysis scenarios.

Chronic wounds, a critical global health challenge, necessitate advanced solutions to address limitations in current care, such as subjective assessments, uncontrolled drug delivery, and poor integration of diagnosis and treatment. This study introduces a "clinic-in-a-dressing" system that combines in situ pH monitoring with dual-responsive (pH/enzyme) drug delivery for chronic wound management. The system integrates hyaluronic acid (HA)-encapsulated vancomycin-loaded zeolitic imidazolate framework (HZV) nanoparticles and fluorescein isothiocyanate (FITC) into a nanofiber-reinforced hydrogel sponge.
