Article Abstract

Bardet-Beidl syndrome (BBS) manifests from genetic mutations encoding for one or more BBS proteins. BBS4 loss impacts olfactory ciliation and odor detection, yet the cellular mechanisms remain unclear. Here, we report that mice exhibit shorter and fewer olfactory sensory neuron (OSN) cilia despite retaining odorant receptor localization. Within OSN cilia, we observed asynchronous rates of IFT-A/B particle movements, indicating miscoordination in IFT complex trafficking. Within the OSN dendritic knob, the basal bodies are dynamic, with incorporation of ectopically expressed centrin-2 and γ-tubulin occurring after nascent ciliogenesis. Importantly, BBS4 loss results in the reduction of basal body numbers separate from cilia loss. Adenoviral expression of BBS4 restored OSN cilia lengths and was sufficient to re-establish odor detection, but failed to rescue ciliary and basal body numbers. Our results yield a model for the plurality of BBS4 functions in OSNs that includes intraciliary and periciliary roles that can explain the loss of cilia and penetrance of ciliopathy phenotypes in olfactory neurons.

Frailty, often linked to sarcopenia, involves reduced muscle strength and mass. While sarcopenia has multiple causes, impaired muscle protein synthesis may contribute. Leucine and resistance training (RT) are anabolic stimuli, but the long-term effects of leucine combined with RT in pre/frail older women remain unclear.

The parasitic protozoan Trypanosoma brucei has a single mitochondrial nucleoid, anchored to the basal body of the flagellum via the tripartite attachment complex (TAC). The detergent-insoluble TAC is essential for mitochondrial genome segregation during cytokinesis. The TAC assembles de novo in a directed way from the probasal body towards the kDNA.

Flaxseed oil contains elevated levels of omega-3 fatty acids (n-3 FA), which have been shown to impact reproductive performance. This study aimed to determine the effects of a flaxseed oil-based supplement on reproductive parameters, feeding behavior, and lipid profile in beef heifers. Sixty Angus and Simmental × Angus heifers (14 months old ± 2 months), blocked by full body weight (BW; 396.

Background: Dry-fermented soybean meal (DFSBM) and wet-fermented soybean meal (WFSBM) were evaluated as alternatives to conventional soybean meal in piglet diets, focusing on growth performance, antioxidant function and fecal microbiome modulation. A total of 225 male piglets (Duroc × (Landrace × Yorkshire)) aged 40 days with a body weight of 13.01 ± 0.

Introduction: This post hoc analysis of an A Toujeo Observational Study (ATOS) aims to evaluate the real-world effectiveness and safety of insulin glargine 300 U/ml (Gla-300) in high-risk subgroups of insulin-naïve people with type 2 diabetes (PwT2D) from multiple geographical regions (Asia, the Middle East, North Africa, Latin America, and Eastern Europe).

Methods: In these post hoc analyses of ATOS, a real-world, 12-month, prospective study included 4422 insulin-naïve adults (age ≥ 18 years) with type 2 diabetes (T2D) uncontrolled (HbA > 7% and ≤ 11%) on one or more oral antidiabetic drugs (OADs) who initiated Gla-300 treatment as per routine practice. Primary and secondary endpoints were studied according to renal impairment (RI) status (without or with) and age group ( View Article and Find Full Text PDF
