Article Abstract

The integrative conjugative element (ICE) Tn of , conferring resistance to tetracycline and chloramphenicol, was found integrated at a 83-bp specific target site (B) located in the gene of the pneumococcal chromosome. PCR analysis of Tn-carrying strains showed evidence of precise excision of Tn from the pneumococcal chromosome with production of (i) circular forms of the ICE in which the ends were joined by a 84-bp sequence (Tn), and (ii) reconstituted chromosomal B. When integrated into the chromosome, Tn was flanked by L, identical to B, and R, identical to Tn. Circular forms of Tn were present at a concentration of 3.8 × 10 copies per chromosome, whereas reconstituted B sites were at 3.0 × 10 copies per chromosome. Deletion of of Tn abolished production of circular forms (<7.1 × 10 copies per chromosome) and was associated to the lack of Tn conjugal transfer suggesting, as expected, that Tn circular form acts as a conjugation intermediate.

We present a supramolecular templating strategy for inducing chirality in hybrid perovskites via confined crystallization within chiral super spaces-nanoconfined, helically ordered cavities formed by the self-assembly of achiral bent-core molecules with chiral additives. Upon removal of the additives, the resulting porous films retain permanent chirality. Quasi-2D hybrid organic-inorganic perovskites crystallized within these templates exhibit distinct chiroptical activity, including mirror-image circular dichroism and circularly polarized light emitting (CPLE), with CPLE dissymmetry factors reaching up to 1.

Mismatch-sensitive DNA hybridization controlled by inchworm-type peptide nucleic acid-PEG conjugates.

The duplex-forming behavior of an inchworm-type PNA-PEG conjugate (i-PPc), engineered for the selective recognition of point mutations in DNA, was assessed through thermodynamic analysis employing UV melting curves and circular dichroism spectroscopy. The i-PPc demonstrated the ability to form stable duplexes exclusively with fully complementary DNA sequences, while no hybridization with single-base mismatched sequences. This binary on/off hybridization behavior was maintained even under physiologically relevant conditions (37 °C), thereby illustrating the exceptional point mutation discrimination capability of i-PPc.
